Abstract: Die vorliegende Arbeit umfasst die Konzeptualisierung und Herstellung eines lokal-wirksamen, kontrolliert freisetzenden Drug Delivery Systems für die Behandlung von Parodontitis. Dafür wurden Chelatkomplexe aus Tetracyclin-derivaten und Fettsäuresalzen hergestellt. Diese Chelatkomplexe stabilisierten den Arzneistoff und ermöglichten die Einarbeitung in PLGA mittels Schmelzextrusion. Die Minocyclin-Magnesiumstearat-Komplexe im molaren Verhältnis 1:2 traten dabei als Favoriten hervor. Die Extrudate boten eine kontrollierte Freisetzung über 42 Tage in einer bioabbaubaren Matrix. Mikrobiologische Untersuchungen zeigten in vitro eine längere Wirksamkeit im Vergleich zu einem Marktpräparat. Nachfolgend wurden die Extrudate durch PEG als Weichmacher und PEG-PLGA als Matrix weiterentwickelt. Dadurch wurden exzellente Flexibilität und mechanische Stabilität erreicht, die eine einfache Handhabung der Extrudate für künftige, klinische Anwendungen ermöglichen werden.
The present work covers the conceptualization and manufacturing of a local acting, controlled release drug delivery system for the treatment of periodontitis. Therefore, chelate complexes consisting of a tetracycline antibiotic and a fatty acid salt were developed. These chelate complexes stabilized the active pharmaceutical ingredient and enabled the incorporation into PLGA matrices via hot melt extrusion. Minocycline-magnesium stearate complexes in a molar ratio of 1:2 emerged as favorites. The extrudates offered a controlled release over 42 days within a biodegradable matrix. Antimicrobial characterization demonstrated prolonged in vitro activity in comparison to a commercial product. Subsequently, the first-generation prototypes were improved through the implementation of PEG 1500 as plasticizer and PEG-PLGA as matrix. Thus, excellent flexibility and mechanical resilience were achieved, which enable an effortless handling of the extrudates for future clinical applications.
