dc.description.abstractDas Tat-Translokationssystem vermittelt in Chloroplasten den Transport vollständig gefalteter Proteine unterschiedlichster Form über die Thylakoidmembran, ohne dass es zu einem Zusammenbrechen des Membranpotenzials kommt. Um den Mechanismus der Translokation untersuchen zu können, soll die Tat-Translokase in künstliche Membransysteme rekonstituiert werden. In dieser Arbeit wurde untersucht, ob sog. Rezeptorkomplexe, die sich aus den Membranproteinen TatB und TatC zusammensetzen, hierfür als Ausgangspunkt genutzt werden können. Betrachtet wurden die Solubilisierung, Charakterisierung, Gewinnung und Rekonstitution von TatBC-Rezeptorkomplexen aus Thylakoiden. Die Solubilisierung führt zu einem schrittweisen Verlust von TatB aus den Komplexen, wobei schließlich ein reines TatC-Oligomer verbleibt. Gebundene Proteine und Calciumionen hemmen diesen Prozess. Es wurde im Folgenden ein Verfahren zur Anreicherung der Komplexe entwickelt und schließlich die Rekonstitution von TatC-Oligomeren in künstliche Membranen gezeigt - ein entscheidender Schritt für die zukünftige Rekonstitution der Tat-Translokase.ger
dc.description.abstractIn chloroplasts, the Tat translocation system mediates the transport of fully folded proteins of various shapes across the thylakoid membrane without collapse of the membrane potential. To be able to study the mechanism of translocation, Tat translocase should be reconstituted into artificial membrane systems. In this work it was investigated whether so-called receptor complexes, which are composed of the membrane proteins TatB and TatC, can be used as a starting point for this purpose. The solubilization, characterization, recovery and reconstitution of TatBC receptor complexes from thylakoids were analyzed. Solubilization leads to a gradual dissociation of TatB from the complexes, ultimately leaving a pure TatC oligomer. Bound proteins and calcium ions inhibit this process. Subsequently, a procedure for enrichment of the complexes was developed and finally reconstitution of TatC oligomers into artificial membranes was demonstrated - a crucial step for future reconstitution of Tat translocase.eng
