The combination of nucleic acid-based amplification techniques with the simple and robust lateral flow technology offers great potential for molecular PoC diagnostics. In this work, Nucleic Acid Lateral Flow Immunoassays (NALFIAs) with practical relevance were developed and investigated with respect to their functionality and performance. General methodological weaknesses and limitations were identified and critically discussed. Approaches to solve these methodological bottlenecks were pointed out and tested for feasibility and practicability. Non-labeled oligonucleotides were identified as valuable tools for the elimination of primer-/probe-dimerization and the improvement of rapid hybridization reactions. Moreover, an innovative, RNaseH2-reporter dependent NALFIA method, that circumvents various methodological limitations and allows accurate analysis, was developed.
