dc.description.abstractDas Flügelfell ist eine invasiv wachsende, stark durchblutete Neoplasie, unklarer Pathogenese, am ehesten limbalen Ursprungs. Risikofaktoren sind tangentiale UV-Strahlung, chronische Mikrotraumata und Entzündungen. Wir untersuchten NGF, seine Vorstufe pro-NGF sowie TNFα hinsichtlich ihrer wachstumssteigernden und migrationsfördernden Effekte an cornealen (HCE) und konjunktivalen (HCjE) Epithelzelllinien. NGF und pro-NGF senkten das Zellwachstum signifikant, wohingegen TNFα das Wachstum förderte. Alle drei Faktoren hatten keinen Einfluss auf die Zellmotilität. Histologische Untersuchungen humaner Proben von Limbus, Horn- und Bindehaut sowie Pterygien zeigten insbesondere epithelial Vorkommen von pro-NGF und TNFα an. An HCE-Zellen konnten alle drei Faktoren nachgewiesen werden, in HCjE nur NGF und TNFα. Erhöhte pterygiale Sekretion von NGF und pro-NGF könnten eine Art körpereigener Abwehrmechanismus darstellen, der das Wachstum des Flügelfells bremsen soll.-
dc.description.abstractA pterygium is an invasive, highly vascularized growth, thought to arise from proliferating limbal epithelial stem cells. Its pathogenesis is uncertain. Environmental factors such as UV-light, chronic irritation, and inflammation are causative factors. We examined NGF, its precursor pro-NGF and TNFα, with regard to cell proliferation and motility effects on corneal (HCE) and conjunctival (HCjE) epithelial cell lines as well as in pterygia. Proliferation of HCE and HCjE was significantly inhibited after stimulation with NGF/pro-NGF whereas TNFα increases cell proliferation. All three factors had no effect on cell motility. Histologic investigation of human samples from limbus, cornea, conjunctiva and primary pterygia revealed presence of pro-NGF and TNFα especially in epithelial cells. In cells all three factors could be visualized in HCE whereas in HCjE only NGF/TNFα was detectable. Increased secretion of NGF/pro-NGF in pterygium may function in blocking growth of the pterygium itself.eng
