The plant pathogenic bacterium Xanthomonas campestris pv. vesicatoria utilizes a type III secretion (T3S) system to translocate effector proteins into plant cells. In the present study, control proteins and conserved components of the T3S system from X. campestris pv. vesicatoria were characterized. The results show that the control protein HpaC induces a switch in the T3S substrate specificity that is mediated by the C-terminal cytoplasmic domain of the conserved HrcU protein. The efficient secretion of effector proteins depends on the general T3S chaperone HpaB, which interacts with effectors and the cytoplasmic ATPase HrcN that dissociates HpaB-effector complexes. The chaperone activity of HpaB is presumably controlled by the translocated regulator HpaA, which binds to HpaB and thus promotes the efficient secretion of extracellular components of the T3S system.eng
