Abstract: Ein möglicher Einfluss von Sexualhormonen & Insulin wird auf die Entstehung von Arthrose vermutet. Die Östrogenrezeptoren ERα & ERβ sowie der Insulinrezeptor IR sollen an humanen Gelenkknorpelzellen auf mRNA- & Proteinebene detektiert werden. Durch verschiedene hormonelle Stimulationen soll ein Zusammenhang der Wirkung zwischen 17β-Östradiol & Insulin aufgezeigt werden. Die immortalisierten Knorpelzelllinien C-28/I2 & T/C-28a2 sowie primäre humane Gelenkknorpelzellen werden in Zellkulturen mit beiden Hormonen stimuliert. Die Hormonrezeptoren werden mit PCR, Western Blot & Immunzytochemie bestimmt. Zusätzlich wird der radioaktiv markierte Prolineinbau in einer Zellkultur untersucht & die Kollagensynthese mittels ELISA bestimmt. Der Nachweis der Rezeptoren gelingt auf mRNA- & Proteinebene. Eine unterschiedlich starke Exprimierung kann in Abhängigkeit von Geschlecht & variierenden Hormonstimulationen teilweise gezeigt werden. Der Prolineinbau & der Kollagengehalt in der Zellkultur geben Anhaltspunkte für eine Interaktion von 17β-Östradiol und Insulin. Diese in vitro nachgewiesenen Zusammenhänge können ein erster Hinweis für tatsächliche Interaktionen in vivo darstellen.
A possible influence of sexual hormones & insulin is expected in the development of osteoarthritis. The estrogen receptors ERα & ERβ as well as the insulin receptor IR are supposed to be detected on mRNA & protein level. A connection between the effect of 17β-estradiol & insulin is supposed to be shown with different hormonal stimulations. The immortalized chondrocyte cell lines C-28/I2 & T/C-28a2 as well as primary human articular chondrocytes in cell cultures are stimulated with both hormones. The hormone receptors are detected with PCR, western blot & immunocytochemistry. Additionally the radioactive marked prolin incorporation is analyzed & the synthesis of collagen is detected with ELISA. The detection of the receptors is successful on mRNA & protein level. A different expression can be shown partially in dependence of sex & various hormonal stimulations. The prolin incorporation & the concentration of collagen in the cell culture assumes an interaction of 17β-estradiol & insulin. These detected connections in vitro can be a first indication for real interactions in vivo.
