Abstract: Sialinsäuren sind ein typischer Bestandteil der N-Glykane der Säugetiere. Sie erhöhen z. B. die Halbwertszeit von Serumproteinen, werden aber mit der Zeit von Sialidasen abgespalten. Infolgedessen werden die Glykoproteine von Asialoglykoproteinrezeptoren der Leber gebunden und aus dem Blutstrom entfernt. In dieser Arbeit wurde die Sialylierung von in CHO-S-Zellen exprimiertem Erythropoietin durch Zugabe der modifizierten Sialinsäurevorläufer N-Pentanoylund N-Propanoylmannosamin zur Zellkultur verändert und massenspektrometrisch nachgewiesen. Die Stabilität der Sialylierung wurde in einem Degradationsassay mit Sialidase A, der durch eine 2D-Gelelektrophorese ausgewertet wurde, überprüft. Es stellte sich heraus, dass die neuartigen Sialinsäuren, insbesondere N-Pentanoylneuraminsäure, im Vergleich zur nativen NAcetylneuraminsäure resistenter gegenüber dem Abbau durch Sialidase A sind, wodurch sich die Halbwertszeit therapeutischer Glykoproteine verlängern ließe.
The terminal positions of N-linked complex glycans in mammals are typically occupied by sialic acids. The presence of sialic acid is crucial for functionality and affects the half-life of glycoproteins. However, glycoproteins in the bloodstream become desialylated over time and are recognized by the asialoglycoprotein receptors via the exposed galactose and targeted for degradation. Non-natural sialic acid precursors can be applied to the cell culture medium to engineer the glycosylation side chains by biochemically introducing new non-natural terminal sialic acids. The sialylation of erythropoietin (EPO) was analyzed. By feeding cells with Npropanoyl or N-pentanoyl mannosamine it was possible to incorporate N-propanoyl or Npentanoyl neuraminic acid in significant amounts into EPO. Using a degradation assay with sialidase A, a higher resistance of EPO to sialidase after incorporation of N-propanoyl and most notably N-pentanoyl neuraminic acid was observed.
