dc.description.abstractZiel der Arbeit war es den Einfluss von Nettoladung und Protein-Protein-Interaktion monoklonaler Antikörper auf ihre physikalische Stabilität, Selbstassoziation und Viskosität in hohen Konzentrationen zu studieren. Nach der Bestimmung der physikalischen Eigenschaften (Nettoladung, iso-elektrischer Punkt, Hydrophobizität) wurden diese in Bezug auf Protein-Protein Interaktionen untersucht (zweiter Virialkoeffizient, A2, mittels statischer- und dynamischer- Lichtstreuung). Es konnte eine Hochdurchsatzmethode für A2 entwickelt werden, die es ermöglichte ca. 600 pharmazeutisch relevante Formulierungen zu testen (IgG1 gegen IgG4, Ionenstärke, pH, Anionen Valenz) und Zusammenhänge zu finden. Schließlich konnte gezeigt werden, dass der bei geringen Konzentrationen gemessene Parameter, A2, genutzt werden kann um Antikörperverhalten in hohen Konzentrationen vorauszusagen (Viskosität, physikalische Stabilität, Trübung). Ein klarer Zusammenhang zwischen Selbstassoziation und A2 war nicht gegeben.-
dc.description.abstractThe purpose of this work was to study how net charge and protein-protein interactions of monoclonal antibodies can be related to their physical protein stability, self-association, and high concentration viscosity behavior. After assessing physical properties (net charge, isoelectric point, zeta potential, hydrophobicity), protein-protein interactions were analyzed (second virial coefficient, A2, by static- and dynamic- light scattering). A high throughput method for A2 could be developed that enabled screening about 600 pharmaceutical relevant buffer systems that allowed studying of various effects (IgG1 vs. IgG4, ionic strength, pH, anion buffer valences) A2. Finally the globally assessed parameter A2 (analyzed at low concentrations) was found to be a predictive parameter for some high concentration protein behavior (viscosity, physical stability, turbidity) but being limited in predicting specific interaction behavior (self-association) effects.eng
