Abstract: KTI11 alias DPH3, kodiert für ein kleines, hoch konserviertes Zink-Finger Protein der Zn-ribbon Klasse. Kti11 interagiert mit mindestens drei verschiedenen Proteinen oder Proteinkomplexen: dem Dph-Komplex, der für die Diphthamid Biosynthese am eukaryotischen Translationsfaktor 2 (eEF2) notwendig ist, dem Elongator-Komplex, der an der wobble Uridin Modifikation eukaryotischer tRNA-Spezies beteiligt ist und Kti13, einem potentiellen Elongator-Regulator. Durch Komplementation einer kti11 Hefe Mutante mit dem Pflanzengen konnten wir nachweisen, dass At2g15910 für ein funktionell konserviertes Protein (AtKti11) kodiert. TAP-Aufreinigungen mit AtKti11 in planta identifizierten den Elongator-Komplex und AtKti13 als mögliche Interaktionspartner. Aus Hefe und E. coli exprimiertes Kti11 zeigte eine Eisen-Bindekapazität. Eine reversible Oxidation von Kti11 deutet auf eine mögliche Funktion als Elektronen-Überträger hin. Der biochemische Reaktionsmechanismus der Elongator-abhängigen tRNA Modifikation ist bis heute ungeklärt, jedoch wird ein radikalischer Mechanismus vermutet, da die Elongator-Untereinheit 3 eine radikal-SAM Domäne trägt. Aufgrund dieser Daten und mit den in dieser Arbeit erzielten Ergebnissen, dass Kti11 Redoxaktivität aufweist, schlagen wir eine mögliche Kti11 Funktion in der Aufrechterhaltung des reduzierten Redoxzustandes der katalytischen Untereinheit 3 vor.
Kti11 alias Dph3, a small, highly conserved Zn-ribbon protein, was found to interact with at least three distinct proteins or protein complexes in yeast: the Dph complex required for synthesis of the diphthamide modification on translation factor eEF2, the Elongator complex involved in modification of wobble nucleosides in eukaryotic tRNAs and Kti13, a protein assumed to be an Elongator regulator. By complementation assays we have shown that plant gene At2g15910 encodes a protein, named AtKti11 that is functionally conserved between yeast and plants. TAP-purification studies in planta identified the Elongator complex and AtKti13 as possible interaction partners. Expressed Kti11 from Yeast and E. coli revealed iron binding capacity. Reversible oxidation of the protein suggests that it might function as an electron carrier. The biochemical reaction of Elongator dependent tRNA modification is still elusive, but a radical based mechanism is predicted, since Elongator subunit Elp3 harbors a radical SAM domain. Since we know that Kti11 is redox active, we hypothesize that Kti11 might function in maintaining the reduced state of catalytic subunit Elp3.
