Abstract: Ochratoxin A (OTA) ist ein Mykotoxin, das mit der Entstehung chronischer Nephropathien in Verbindung gebracht wird. Es wird von Schimmelpilzen synthetisiert und ist ubiquitär in Lebens- und Futtermittel verbreitet. Die zu Grunde liegenden molekularen Mechanismen, die nach einer Exposition mit relevanten OTA-Konzentrationen zu fibrotischen Nephropathien führen können, sind jedoch weitgehend unbekannt. Durch die im Rahmen der Dissertation erzielten Ergebnisse kann die pro-fibrotische Wirkung von OTA dadurch erklärt werden, dass OTA die Expression von einer neuartigen, langen nicht-kodierenden (lncWISP1) induziert. LncWISP1 wiederum stellt alternative Bindestellen für die anti-fibrotisch wirkende miR-29b bereit, wodurch miR-29b nicht mehr an ihre eigentlichen Zieltranskripte (u.a. diverse Kollagene) bindet, so dass deren Expression moduliert wird. Damit begünstigt lncWISP1 und somit auch OTA indirekt fibrotische Prozesse, indem die anti-fibrotische miR-29b in ihrer Wirkung unterdrückt wird.
Ochratoxin A (OTA) is a mycotoxin suspected to cause chronic nephropathies. It is synthesized by different fungi and is an ubiquitous contaminant of food and feed. The underlying molecular mechanisms that are responsible for the OTA-induced fibrotic nephropathies are still unknown. The results of this PhD thesis suggest that the pro-fibrotic effects of OTA can be explained by the expression of a novel, noncoding RNA (lncWISP1), which provides alternative binding sites for the anti-fibrotic miR-29b. In consequence, miR-29b can´t bind to its normal target mRNA (e.g. collagens) leading to a changed expression of this target transcripts. Thus, lncWISP1 and therefore OTA can promote fibrotic tissue alterations indirectly by inhibiting the effects of the anti-fibrotic miR-29b.
