dc.description.abstractHelicobacter pylori ist ein gramnegatives Epsilon Proteobakterium, welches den Magen von ca. 50% der Bevölkerung besiedelt. In Folge einer Infektion kann es zur Entwicklung von Magengeschwüren kommen. H. pylori kommt in verschiedenen Morphologien vor. Während die spirale Morphologie motil und infektiös ist, zeigen die coccoiden Formen keine oder sehr eingeschränkte Infektiosität. In dieser Arbeit wurde eine Methode zur Quantitativen Proteomuntersuchung von H. pylori Stamm 26695 etabliert, um Unterschiede zwischen der spiralen und der vitalen coccoiden Morphologie zu untersuchen. Hierfür wurde zunächst die Abdeckung von niedermolekularen Proteinen optimiert. Des Weiteren wurde eine proteogenomische Studie des Stamms 26695 durchgeführt, um die Datenbankqualität zu verbessern. Hierdurch konnten vier bisher nicht annotierte Proteine detektiert werden. Des Weiteren wurden sechs Proteinannotationen korrigiert und 63 Signalpeptidaseschnittstellen identifiziert. In der abschließenden Studie wurde die stabile Isotopenmarkierung durch Aminosäuren in Zellkultur (engl. stable isotope labeling by amino acids in cell culture, SILAC) für H. pylori Stamm 26695 etabliert. Mittels SILAC wurden in einer Studie Änderungen im Proteom zwischen der spiralen und vitalen coccoiden Morphologie relativ quantifiziert. Hierbei wurde gezeigt, dass Proteine, die essentiell für Chemotaxis und Pathogenese sind, in coccoiden Zellen schwächer exprimiert werden. Die etablierte Methode zur relativen Quantifizierung des Proteoms von H. pylori eröffnet neue Möglichkeiten, wie beispielsweise die Untersuchung der Wirkung von verschiedenen Antibiotika auf Proteinebene.-
dc.description.abstractHelicobacter pylori is a Gram-negative epsilon proteobacterium that colonizes the stomach of approximately 50% of the human population. Infections can cause the development of gastric ulcer diseases. H. pylori occurs in different morphologies. The spiral morphology is motile and infectious whereas coccoid forms show no or strongly reduced infectivity. In this thesis, a method for relative quantification of H. pylori strain 26695 proteome was developed to investigate changes in the proteome of the spiral and the vital coccoid morphology. Therefore, the coverage of low molecular weight proteins was optimized. Additionally, a proteogenomic study was carried out to improve the database quality of H. pylori strain 26695. In this study, four previously not annotated proteins were discovered. Additionally, six protein annotations could be corrected. Moreover, 63 signal peptidase cleavage sites were identified. In the final study, stable isotope labeling by amino acids in cell culture (SILAC) was established for H. pylori strain 26695. This technique was used to perform relative protein quantification between the spiral and the vital coccoid morphology of H. pylori. Essential proteins involved in chemotaxis and infectivity were found to be down-regulated in coccoid cells. The established method provides the opportunity for new study designs, such as the study of the influence of antibiotics on H. pylori on protein level.eng
