Abstract: Das erste Ziel dieser Arbeit war, zu beurteilen, ob haploide Drohnenlarven als Studienorganismen für QTL-Studien zur Krankheitstoleranz bei Honigbienen geeignet sind. Dafür verglichen wir Honigbienendrohnen mit -arbeiterinnen hinsichtlich ihres Überlebens nach Infektion mit dem bakteriellen Pathogen Paenibacillus larvae, das Amerikanische Faulbrut (AFB) verursacht. Wir schlussfolgerten, dass Drohnen Arbeiterinnen in AFB-Resistenz-Studien ablösen können, und ermittelten eine Abschätzung der letalen Bakterienzellzahlen für Arbeiterinnen und Drohnen. Das zweite Ziel dieser Arbeit war, haploide Drohnen zu nutzen, um QTL für die larvale Resistenz gegen zwei Hauptschädlinge der Honigbiene, nämlich AFB und die Varroa-Milbe, zu finden. Für die larvale AFB-Resistenz identifizierten wir einen signifikanten und drei suggestive QTL sowie eine signifikante epistatische Interaktion und für die larvale Varroa-Resistenz einen signifikanten sowie zwei suggestive QTL mit epistatischen Interaktionen.
The first aim of this thesis was to evaluate haploid drones as study organisms for QTL studies on disease tolerance in honeybees. We therefore compared honeybee drones and workers in their prepupal survival after an infection with the bacterial pathogen Paenibacillus larvae causing American Foulbrood (AFB). We concluded that honeybee drones may replace workers in studies on individual AFB resistance and provided an estimation of lethal bacterial cell numbers in both worker and drone larvae. The second aim of this thesis was to use haploid drones to identify major loci affecting larval resistance against two major pests of honeybees, namely AFB and the Varroa mite. In the case of larval AFB resistance we identified one significant and three suggestive QTL as well as one significant epistatic interaction. In the case of larval Varroa resistance we identified one significant and two suggestive QTL for larval suppression of mite reproduction with strong epistatic interactions.
