Abstract: Zur die Identifizierung möglicher Virulenzfaktoren des Gerstenpathogens Rhynchosporium commune wurden einzelne Kandidatengene (Pektinmethylesterase: PME1; mutmaßliches RXLR-Effektorprotein: RxP1; Polyketidsynthasen: PKS1, PKS2, PKS3) deletiert. Die Deletionsmutanten DPME1, DRxP1, DPKS1 und DPKS3 zeigten ein verstärktes Wachstum in planta, das außer bei DPME1 auch mit schnellerer Symptomausbildung korrelierte. Dagegen wird das Pilzwachstum in planta durch die Deletion von PKS2 kaum beeinflusst. Die Kulturfiltrate von DPKS1 und DPKS3 sind durch Fehlen bzw. die mengenmäßige Reduktion einer Substanz mit der Summenformel C15H14O6 charakterisiert. Erste Untersuchungen zur Funktion dieser Verbindung deuten auf eine autoinhibitorische Wirkung hin. Nach den im Rahmen dieser Dissertation erlangten Erkenntnissen kann für PME1, RxP1, das PKS2-Produkt sowie die Verbindung C15H14O6 eine Rolle in der Pflanze-Pathogen-Interaktion postuliert werden.
To identify putative virulence factors of the barley pathogen Rhynchosporium commune single candidate genes (pectin methyl esterase: PME1; putative RXLR effector protein: RxP1; polyketide synthases: PKS1, PKS2 and PKS3) were deleted. DPME1, DRxP1, DPKS1 and DPKS3 showed enhanced growth in planta that except for DPME1 correlated with faster symptom development. In contrast fungal growth in planta is hardly affected by PKS2 deletion. Culture media of DPKS1 and DPKS3 are characterized by the lack and reduced amounts, respectively, of a compound with the formula C15H14O6. Preliminary data indicate an autoinhibitory function of this compound. The results of this thesis suggest a role of PME1 and RxP1, of the product of PKS2 and of the compound C15H14O6 in the plant-pathogen interaction.
