Abstract: Für Untersuchungen zum Replikationszyklus von Plusstrang-RNA-Pflanzenviren und Virus-Wirt-Interaktionen sind in vitro-Systeme von Vorteil, die eine synchronisierte Translation und Replikation des viralen RNA-Genoms ermöglichen. In der vorliegenden Arbeit wurde für Tomato bushy stunt virus (TBSV) ein authentisches in vitro-System, basierend auf Nicotiana tabacum Zelllysat, etabliert. Die Charakterisierung der TBSV-Replikation in diesem in vitro-System zeigte, dass sich ein aktiver Replikase-Komplex mit hoher Matrizen-Spezifität bildet und die Replikation ohne simultane Translation möglich ist. Mutationen in einem replikationsregulatorischen RNA-Element bestätigten dessen Bedeutung für die Replikation in einem Schritt, der der (-)-Strang-Synthese vorausgeht. Viruspartikel-Assemblierung war in diesem Translations-/Replikations-System nicht nachweisbar. Die RNA-abhängige RNA-Polymerase des TBSV wurde rekombinant gereinigt und zeigte Nukleotidyltransferase-Aktivität in vitro.
In vitro systems which allow for synchronized translation and replication are beneficial for investigations of the replication cycle of (+)-strand RNA viruses and its interactions with a host. In this study an authentic in vitro system based on Nicotiana tabacum cell lysate was established for the plant pathogen Tomato bushy stunt virus (TBSV). Characterizing the TBSV replication in this system it was shown that an active replication complex is formed with high specificity for its template. The replication does not depend on simultaneous translation, demonstrating that both processes are not functionally linked in this system. Mutational analysis of the TBSV RNA confirmed the importance of a replication regulating element for the RNA replication in a step preceding the (-)-strand synthesis. Virus particles did not assemble in the in vitro translation/replication system. The TBSV RNA-dependent RNA polymerase was recombinantly purified and showed nucleotidyltransferase activity in vitro.
