dc.description.abstract | In der vorliegenden Arbeit wurde eine effiziente, leicht zu handhabende Screening-Methode zur Bestimmung von Vitamin D (Ergosterol, 7-Dehydrocholesterol, Cholecalciferol, Ergocalciferol, 25-Hydroxycholecalciferol, 25-Hydroxyergocalciferol, 1α,25-Dihydroxycholecalciferol, 1α,25-Dihydroxyergocalciferol, (24R)-24,25-Dihydroxycholecalciferol) in Lebensmitteln entwickelt, hausintern validiert und angewendet. Die Untersuchung von über 1.500 Realproben belegte deren Eignung für die Routineanalytik. Lebensmittelproben wurden nach Zugabe von isotopenmarkierten Standards methanolisch hydrolysiert und mittels kombinierter Festphasenextraktion (Normal- und Reversed-Phase) aufgereinigt und aufkonzentriert. Die Identifizierung und Quantifizierung erfolgte mittels LC-MS/MS im scheduled Multiple Reaction Monitoring-Modus (ESI, positiv). | - |
dc.description.abstract | In the present work, an efficient, easy-to-use screening method for the determination of vitamin D (ergosterol, 7-dehydrocholesterol, cholecalciferol, ergocalciferol, 25-hydroxycholecalciferol, 25-hydroxyergocalciferol, 1α,25-dihydroxycholecalciferol, 1α,25-dihydroxyergocalciferol, (24R)-24,25-dihydroxycholecalciferol) in food was developed, in-house validated and applied. The suitability for routine analysis was proven by the study of more than 1,500 real samples. After spiking with isotope-labeled internal standards, food samples were saponified, purified and concentrated by combined solid-phase extractions (normal- and reversed-phase). Identification and quantification were performed by liquid chromatography-tandem mass spectrometry using scheduled multiple reaction monitoring mode (ESI, positive). | eng |
