Abstract: Diese Studie wurde an einem Septic Shock Modell an Ratten durchgeführt. Es erfolgte die Einteilung in drei Versuchsgruppen: Sham-Gruppe, LPS Shock + Vehicle-Gruppe, LPS Shock + Medikament-Gruppe. Das jeweilige Medikament wurde einen Tag vor der LPS-Gabe intraperitoneal verabreicht. Am Versuchstag wurden die Tiere mit Pentobarbitali.p. betäubt und durch LPS in einen septischen Schock versetzt. Nach dem Versterben der Tiere erfolgte die Entnahme von Herz, Lunge, Leber, Nieren. Von diesen Geweben wurden Western Blots und histologische Präparate angefertigt sowie die Myeloperoxidaseaktivität bestimmt. Für LPS Shock + Pioglitazon-Gruppe ergab sich eine Verdopplung der Überlebenszeit gegenüber der LPS Shock + Vehicle-Gruppe. In den histologischen Präparaten war in allen vier Organen auch eine geringere Schädigung des Gewebes sichtbar. Die Analyse der MPO-Aktivität und der Proteinexpression lässt einen protektiven Effekt von Pioglitazon im septischen Schock vermuten. Die Gabe von Ranolazin dagegen hatte weder einen positiven Einfluss auf das Überleben noch besserten sich die Entzündungsparameter. Die Gewebeschädigung konnte lediglich minimal verringert werden. Insgesamt ließ sich kein protektiver Effekt von Ranolazin im septischen Schock nachweisen.
This study was performed on a septic shock model with Wistar-rats. All animals were assigned to one of the following groups: sham-group, LPS shock + vehicle-group, LPS shock + medicine-group. The medicine was given intraperitoneal one day before LPS-treatment. Before inducing the septic shock with LPS the animals were anaesthesized with pentobarbital. After dying the heart, lungs, lever, kidneys were removed. From these tissuses western blots and histological preparations were performed and the myeloperoxidase-acitivity was measured. Animals of the LPS shock + piogliatzon-group lived twice as long as the LPS shock + vehicle-group. A lower damage of all tissues was visible in the histological preparations. The measurement of the myeloperoxidase-activity supposed a protective effect of pioglitazon on the septic shock. Pretreatment with ranolazin influenced neither the survival time nor the markers of inflammation. The damage of all tissue decreased insignificantly. Summing up no protective effect of ranolazin in the LPS-induced septic shock was verified.
