dc.description.abstractDer Transfer von Polynukleotiden mit Hilfe von kationischen Lipiden ist eine vielverspreschende Methode für Anwendungen in der Medizin und ein wichtiges Werkzeug in der Grundlagenforschung. Das zunehmende Interesse auf diesem Gebiet gilt der Effektivitätssteigerung des Polynukleotidtransfers mit kationischen Lipiden und der Aufklärung grundlegender Prozesse der Polynukleotidkomplexierung und Freisetzung. Im Rahmen dieser Arbeit wurde eine Substanzbibliothek von kationischen Lipiden (Malonamide der 2. Generation) synthetisiert, welche systematische Strukturvariationen sowohl im lipophilen als auch im hydrophilen Molekülteil aufweisen um Struktur/Effektivitäts-Beziehungen aufstellen zu können. Weiterhin wurden Derivate mit Galactoseeinheiten als Erkennungsstrukturen für die Zelltargetierung dargestellt. Wässrige Dispersionen der Lipide wurden mit Hilfe der DSC untersucht, sowie die Mizellbildung einiger Vertreter der neuen Lipide aufgeklärt. Weiterhin wurde die DNA-Komplexierung mit ausgewählten Lipiden mit einer Vielzahl physikochemischer Methoden (DSC, ITC, DLS, Partikelladung, SAXS, TEM) charakterisiert.-
dc.description.abstractThe cationic lipid mediated polynucleotide transfer is a promising method for applications in medicine and an important tool in basic research. The growing interest in this field of research focuses on the increase of efficiency of lipid mediated polynucleotide transfer and the investigation of the complex formation process between cationic lipids and polynucleotides. This work describes the synthesis of a substance library of cationic lipids (malonic acid diamides of the second generation) with systematic structure variations in the lipophilic and the hydrophilic region of the molecules to gain structure/effectivity-relationships. Furthermore, derivatives with galactose moieties in the head group as recognition structures were synthesized. Aqueous dispersions of the lipids were investigated by DSC and the micelle formation of selected lipids was studied. Finally, the DNA complexation of selected lipids was investigated by numerous methods (DSC, ITC, DLS, particle charge, SAXS, TEM).eng
