Abstract: Die chemische Synthese von komplexen, niedermolekularen Kohlenstoffverbindungen ist oft schwierig und mit hohem finanziellen sowie zeitlichen Aufwand verbunden. Die enzymatische Synthese von Feinchemikalien zur Anwendung im Forschungs- und Entwicklungsbereich sowie für Additive in der Lebensmittel- und Duftstoffindustrie, gewinnt deshalb an Bedeutung. Aus diesem Grund sind vor allem alkylierende Enzyme von großer Bedeutung, die chemisch anspruchsvolle C-C-Verknüpfungen katalysieren können. Neben dem Methyltransfer sind hierbei vor allem prenylierende Enzyme von großem Interesse. Daher wurde anhand von zwei Prenyltransferasen (PT) und einer Terpensynthase (TS) die biokatalytische Synthese artifizieller Terpenoide mit synthetischen Prenyldiphsophaten gezeigt. Dabei ist es unter anderem gelungen mit einer thermostabilen PT Chloroprendiphosphat-Oligomere zu biosynthetisieren. Zudem ist die Bildung artifizieller, zyklischer Mono- und Sesquiterpenoide durch eine Terpensynthase gelungen.
The chemical synthesis of complex and low-molecular-weight carbon compounds is difficult and means an increased financial as well as temporal effort. The enzymatic syntheses of fine-chemicals for the application in research and development as well as for additives in food and fragrance industries become more important. For that account, alkylating enzymes are of high interest because they catalyze chemical demanding C-C-bond formations. Besides the methyl transfer, prenylating enzymes are of peculiar interest. Hence, the catalytic ability of two prenyl transferases (PTs) and a terpene synthase for the synthesis of artificial terpenoids was analyzed with synthetic prenyl diphosphate substrates. Among others, the formation of chloroprene diphosphate oligomers by a thermo-stable PT could be shown as well as the biocatalytic formation of new mono- and sesquiterpenoids by a terpene synthase.
