Abstract: Ziel dieser Arbeit war es die Wirkung des Polyvitamin-B Präparates Polybion N® (Merck, Deutschland) auf die Entwicklung des sekundären Gaumens bei einem hereditär spaltbelasteten Mäusestamm (A/WySn) zu untersuchen. Der A/WySn-Mäusestamm weist eine hohe hereditäre Spaltbildungsrate von 20-44% auf und ist gegenüber teratogenen Einflüssen relativ resistent (Juriloff et al., 1982, Schubert et al., 2002). Die Präparation der Organkulturen erfolgte vor der physiologischen Entwicklung des sekundären Gaumens am 14.8 Tag p.c. und der Entwicklungsstand wurde vor und nach der Inkubation (72 h) bestimmt (Al Obaidi et al., 1995) und statistisch ausgewertet. 89% der Gaumenplattenpaare der A/WySn-Maus wiesen unter Standardbedingungen in der Organkultur nach 72 Stunden kein Wachstum auf. Bei Substitution des Kulturmediums mit Polybion N® konnte jedoch ab einer Medienkonzentration von 0,1% ein signifikantes Wachstum der Gaumenplatten (p0,1%=0,000; p1,0%=0,000) beobachtet werden. Die postive Wirkung der klinischen Prophylaxe mit Poly B-Vitaminen konnte in der Organkultur des sekundären Gaumens der A/WySn-Maus bestätigt werden. Durch die direkte Beeinflussung der Strukturen des sekundären Gaumens kam es zu einer signifikanten Entwicklung dieser Strukturen.
This study analyses the direct influence of vitamin B-complex supplements (Polybion®N, Merck Pharma GmbH, Germany) on secondary palate development in palate organ cultures of A/WySnJmice via the supporting media. The inbred strain of A/WySnJ-mice shows a high spontaneous, genetically determined clefting rate of 20-44%. 177 A/WySnJ-fetuses were micro-dissected on the 14.3 GD (gestational day) before palatal fusion takes place. Palatal organ-cultures were prepared and incubated in chemically defined serum-free medium with different concentrations (0.1%, 1.0%) of the vitamin B complex Polybion®N for 72 hours. Palatial development was analysed microscopically according to the six step visual scale of Al-Obaidi (1) that describes the approximation of the palatal shelves during development. At the beginning of the experiment (14.3 GD) the palatal development of all specimens used for the in vitro organ culture showed a clear approach of palatal shelves at stage II. 72 hour after in-vitro cultivation the palatal shelves of the organ-cultures supplemented with vitamin B complex showed a significant growth (p0.1%=0.00017, p1.0%=0.00078) while the untreated control group remained at the initial developmental stage II (p control=0.291). The results of this in vitro study suggest a significant positive influence of vitamin B supplementation on the palatal shelves development in the organ-culture. Further studies will focus on the vitamin B-concentration in the amniotic fluid of mother mice with or without clefts in their offspring.
