Abstract: In höheren Pflanzen gibt es Myosine der Klasse VIII und XI. Es gibt Hinweise, dass die Myosinen der Klasse VIII Funktionen bei der Zell-Zell-Interaktion und der Zellteilung haben. Die Myosine der Klasse XI sind sehr wahrscheinlich für die Entstehung der Cytoplasmaströmung zuständig. Ein Ziel war die Cargo-Spezifität der Arabidopsis thaliana-Klasse-XI-Myosine in A. th. zu untersuchen. Dazu wurden sogenannte tail-Konstrukte aller Klasse-XI-Myosine erstellt. Diese wurden eingesetzt, um die subzelluläre Lokalisierung zu untersuchen und um transgene A. th.-Pflanzen zu generieren. Den Konstrukten der Myosine MyaI, XI-D und XI-I konnten mit geeigneten Markern Oleosomen, Golgi bzw. Tubulin als Cargo zugeordnet werden. Die Generierung transgener A. th.-Pflanzen war erfolgreich. Vier der untersuchten Pflanzenlinien zeigten vesikuläre Strukturen markiert. Weitere Untersuchungen an den transgenen A. th.-Pflanzen lieferten Hinweise, dass eine Gewebsspezifität der Myosin-Cargo-Interaktion vorliegt.
In higher plants there are myosins of class VIII & XI. There are indications that myosins of class VIII have functions in cell-cell interaction and cell division. Class XI myosins are very likely responsible for the formation of cytoplasmic streaming. One aim was to study the cargo specificity of Arabidopsis thaliana class XI myosins in A. th.. For this purpose so-called tail-constructs of all class XI myosins were created. These were used to examine the subcellular localization & taken to generate transgenic A. th.-plants. The constructs of the myosins MyaI, XI-D & XI- I could be allocated with oleosomes, Golgi or tubulin as cargo. The generation of transgenic A. th.-plants was successful. Four of the studied plant lines showed vesicular structures marked. Further studies on the transgenic A. th.-plants provided evidence that a tissue specificity of the myosin cargo interaction exists.
