dc.description.abstractIn dieser Arbeit sollen die Effekte der Transplantation verschiedener Zelltypen (ES-Kardiomyozyten sowie Myoblasten der Skelettmuskulatur) auf das Myokardgewebe im Herzinfarktmodell der Maus echokardiographisch und histologisch untersucht werden. Eine Woche nach Induktion eines Myokardinfarktes erfolgt die Zelltransplantation (bzw. Injektion von PBS als Kontrollgruppe). Die Transplantationen sowohl der Myoblasten als auch der Kardiomyozyten üben positive Effekte auf die Funktion und Morphologie des geschädigten Myokardgewebes aus. Die Myoblasten-Tx führt hier eher zu einer Reduktion der Infarktexpansion, wohingegen die Kardiomyozyten-Tx einen günstigen Einfluss auf das Remodelling hat. Da 28 Tage nach Tx keine relevanten Mengen an funktionell integrierten Zellen nachweisbar sind, muss man davon ausgehen, dass die Wirkung der transplantierten Zellen weniger durch eine direkte funktionelle Integration, sondern eher durch die Freisetzung von Zytokinen hervorgerufen wird.-
dc.description.abstractA growing number of studies are reporting beneficial effects of the cell-transplantation into diseased hearts after myocardial infarction. We have compared the effects exerted by skeletal myoblasts and ESC-derived cardiomyocytes after transplantation into infarcted mouse hearts (cell-transplantation 1 week after infarction; injection of PBS as a control-group) by echocardiographic follow-up and histological analysis. Within the positive effects of the transplanted cells, the myoblasts are more likely to reduce the expansion of the infarction, the cardiomyocytes do have more influence on the remodelling. We postulate that the beneficial effects of the transplantation of myoblasts and cardiomyocytes on heart function and morphology only partially (if at all) depend on the integration of transplanted cells into the myocardium but do depend on the release of a complex blend of cardioactive cytokines.eng
