dc.description.abstractDas Phytohormon Auxin (Indolessigsäure oder IAA) reguliert eine Vielzahl von pflanzlichen Wachstums- und Entwicklungsprozessen über eine hierachische Kontrolle der Genexpression. Zwei Klassen von interagierenden Transkriptionsfaktoren sind für die Expression primärer Zielgene wichtig: die Auxin Response Factors (ARF) und Auxin/IAA-induzierbaren Repressoren (AUX/IAA).In dieser Arbeit wurde die Struktur der C-terminalen Interaktionsdomäne eines AUX/IAA Proteins, PsIAA4 aus der Erbse (Pisumsativum), mit Hilfe der NMR Spektroskopie aufgeklärt, welche Homologien zur Ubiquitin-ähnlichen Phox/Bem1p Domäne (PB1) aufweist. Diese PsIAA4 Domäne verfügt über zwei separate, gegensätzlich geladene und orientierte Oberflächenbereiche, die eine Kopf-Schwanz Multimerisierung über elektrostatische Wechselwirkungen ermöglichen. Unsere Studie liefert einen Beitrag zur detaillierten Aufklärung der molekularen Determinanten, welche die Spezifität der Interaktion von ARF und AUX/IAA Proteinen bestimmen.-
dc.description.abstractThe plant hormone auxin, is a versatile small molecule that is essential during the entire plant life cycle and regulates numerous growth and development processes, mainly via hierarchical control of gene expression. Two main classes of transcription factors (TF) involved in primary response are the AUXIN/INDOLE-3-ACETIC ACID (AUX/IAA)-inducible repressors and the AUXIN RESPONSE FACTOR. In this research work, the solution NMR structure of C-terminal domains III and IV of the AUX/IAA protein PsIAA4 from pea (Pisumsativum) revealed a ubiquitin-like β-grasp fold with homologies to the Phox& Bem1p (PB1) domain. This domain features two distinct surface patches of oppositely charged amino acid residues, mediating front-to-back multimerization via electrostatic interactions which was analysed in vitro and in vivo. Our study provides an impetus for elucidating the molecular determinants that confer specificity to complex protein–protein interaction circuits between AUX/IAA and ARF families.eng
