dc.description.abstractAvrBs3 ist ein Transkriptionsaktivator derTALE Proteinfamilie. TALEs besitzen eine modulare DNA-Bindedomäne, die sich aus einer variierendenAnzahl von 34 AS-Wiederholungen (repeats) zusammensetzt.Jeder repeat vermittelt die spezifische Bindung eines Basenpaares, wobei die Spezifität durch die AS an Position 12 und 13 (repeat-variable diresidue - RVD) definiert wird. In dieser Arbeit wurde die Dimerisierung von AvrBs3 sowie dessen Spezifität für Thymin am 5` Beginn der RVD-definierten DNA-Zielsequenz (T0) analysiert. AvrBs3 bildet Homo-Oligomere durch die Ausbildung von Disulfidbrücken zwischen den Cysteinen der repeat-Region in vitroaus, welche essenziell für die Aktivität und DNA-Bindespezifität von AvrBs3 in planta sind. Die N-terminale Region (NTR), insbesondere ein Tryptophan im degenerierten repeat -1 (W232), begünstigt über die Interaktion mit T0 die Bindung der repeats an die DNA-Zielsequenz. Die Daten führten zu einem zwei-Schritt Modell der TALE-DNA Interaktion: (i) “Suchvorgang“ durch das Gleiten mit einer lockeren Konformation der repeat-Region entlang der DNA und (ii) Bindung der spezifischen DNA-Zielsequenz unter Komprimierung der repeats.-
dc.description.abstractAvrBs3 is a transcription activator of the TALE protein family. TALEs possess a modular DNA-binding domain composed of a varying number of tandem arranged 34 amino acid repeats. Every repeat binds one base pair, in which the specificity is defined by the amino acids at position 12 and 13 (repeat-variable diresidue - RVD). Here, the dimerization of AvrBs3 and its specificity for a thymine at position zero of the RVD-defined target sequence (T0) was analyzed.AvrBs3 forms in vitrohomo-oligomers via disulfide bridges between cysteine residues of the repeat-region, which are additionally essential for the activity and DNA-binding specificity of AvrBs3 in planta. The N-terminal region (NTR),especially the tryptophan in the degenerated repeat 1 (W232), facilitates via the interaction with T0 the binding of the repeat region to the DNA target sequence.This study suggests a two-state model of the TALE-DNA-binding mode:(i)a sliding scan-process of TALEsalong the DNA-strand with a loose conformation of the repeat-region and (ii)a binding-event which leads to a compressed conformation of the repeats.eng
