dc.description.abstractPAR2 ist ein GPCR, der durch Serinproteasen über einen proteolytischen Mechanismus aktiviert wird. PAR2 wird verstärkt in isoliertem, malignem Gewebe exprimiert und gilt als potenzieller Modulator in neoplastischen Erkrankungen. Alle bis dato gezeigten Inhibitoren sind eingeschränkt in ihrer Wirkstärke und Selektivität. Die Arbeit behandelt die Charakterisierung des Naturstoffs Teleocidin A2 als neuer Kandidat für die Inhibition PAR2 stimulierter Signalwege und die daraus resultierende pharmakologische Evaluierung natürlicher Derivate der Substanzklasse sowie neu synthetisierter Indolactam V Derivate anhand zellbasierter Assays. Teleocidin A2 blockierte die PAR2 vermittelte intrazelluläre Mobilisierung von Ca2+ in Brust und Lungenkarzinomzellen potent (IC50 14-26 nM) und selektiv. Teleocidin A2 unterdrückte die PAR2 stimulierte Tumorzellmigration. Drei neue Indolactam V Derivate inhibierten die PAR2 abhängige Ca2+ Freisetzung mit equipotenter Wirkung verglichen mit Teleocidin A2. Im Gegensatz zu den Naturstoffverbindungen, die aus der Literatur als Proteinkinase C Aktivatoren bekannt sind, zeigten die Derivate eine stark reduzierte PKC Aktivierung.-
dc.description.abstractPAR2 is a GPCR, activated by serine protease induced proteolytic cleavage. PAR2 is found to be overexpressed in isolated malignant tissue and was introduced as a modulator responsible for neoplastic disease. All known inhibitors to date are strongly limited in potency and selectivity. The present study focused on the characterization of the natural metabolite teleocidin A2 as a new candidate for inhibition of PAR2 regulated signaling pathways and the pharmacological evaluation of additional natural teleocidin derivatives and newly synthesized Indolactam V derivatives, with the use of cell-based assay systems. Teleocidin A2 potently inhibited PAR2 (IC50 14-26 nM) induced Ca2+ mobilization in breast and lung carcinoma cell lines rather than Ca2+ mobilization of other members of Gq-coupled GPCRs. Teleocidin A2 suppressed PAR2 stimulated MDA-MB 231 cancer cell migration. Three newly synthesized Indolactam V derivatives blocked PAR2 stimulated Ca2+ release dependent on their structure and proved to be equipotent to teleocidin A2. In contrast to teleocidines, known as potent activators of protein kinase C, they showed a reduced activity in stimulating PKC.eng
