dc.description.abstractStreptomycescoelicolor A3(2) zählt zu den hoch GC-reichen grampositiven Actinobacteria und ist bekannt für seinen komplexen Lebenszyklus, der vegetatives Substratmyzel, Luftmyzel und Sporen umfasst. Als obligater Aerobier nutzt das Bakterium Sauerstoff als terminalen Elektronenakzeptor. Zur Energiekonservierung unter aeroben Bedingungen werden die Elektronen über eine bislang wenig erforschte Atmungskette transferiert, die mit multiplen Cytochrom-Oxidasen endet. Obwohl S. coelicolor zum Wachstum O2 benötigt, enthält das Genom 3 Operone für respiratorische Nitratreduktasen (Nars). Damit kann S. coelicolor mit Nitrat atmen, wenn Sauerstoff nicht verfügbar ist. Der regulatorische Switch zwischen Sauerstoff- und Nitratatmung ist Gegenstand der vorliegenden Dissertation. Die Dissertation zeigt neue Aspekte zur aeroben Atmung und die Identifizierung einer neuen Sporen-spezifischenOxidase. Darüber hinaus werden alle dreiNars im Kontext des Lebenszyklus charakterisiert und eine putative Verbindung zur aeroben Atmung entdeckt.-
dc.description.abstractStreptomycescoelicolorA3(2) belongs to the high-GC gram-positive actinobacteria and is characterized by undergoing a complex life-cycle, includingvegetative hyphae, aerial hyphae and spores. As an obligate aerobeS. coelicolorusesaerobic respiration for growth, butstudiesin this area arelimited. Theaerobic respiratorychain terminates with multiple cytochrome oxidases. Despite requiring oxygen for growth, the genome of S. coelicolor habours three operons encoding respiratory nitrate reductases (Nars). Thus,S. coelicolor is able to respire with nitrate when oxygen is not available. The regulatory switch between aerobic respiration and nitrate respiration during the lifecycleis the subject of this dissertation. This work reveals new aspects regarding aerobic respiration, identifying a new spore-specific oxidase.Moreover,all three Nars were analyzed in the context of the developmental programand a putative link between Nar1 and components of the aerobic respiratory chain in spores was established.eng
