Abstract: Die Organisation von amphiphilen Polymerkonjugaten, welche ein β-Turn Element besitzen, wurde in Lipidmembranen untersucht. Die Konjugate bestehen aus ein bzw. zwei hydrophoben Polyisobutylen(PIB)-Ketten, welche am selben hydrophilen β-Turn Element (BTD) angeknüpft sind. Die definierte Anbringung der hydrophoben PIB-Ketten am β-Turn Element erfolgte mittels 1) Amidierungsreaktion und 2) Azid/Alkin-„Klick“-Reaktion. Die Reinheit der synthetisierten Polymerkonjugate wurde mittels NMR Spektroskopie, GPC, MALDI-ToF MS und LC/MALDI-ToF MS überprüft. Das Verhalten der Polymerkonjugate in Lipidmembranen (DPPC) wurde untersucht a) in Monoschichten mittels Kompressions-Isothermen, gekoppelt mit Epifluoreszenz-Mikroskopie und Rasterkraftmikroskopie, und b) in Doppelschichten(GUVs) mittels Konfokalmikroskopie. Es konnte gezeigt werden, dass beide Polymerkonjugate zwar ähnliche Membranwechselwirkungen zeigen, jedoch die „2-Armverbindung“ besser mischbar mit den Lipiden ist.
The organization of amphiphilic polymer conjugates containing a β-turn element was investigated in lipid membranes. These conjugates were based on one or two hydrophobic polyisobutylene (PIB) chains, which are connected onto the same hydrophilic β-turn element (BTD). The defined linkage of the hydrophobic PIB chains onto the β-turn element was accomplished using 1) the amidation reaction and 2) the azide/alkyne-“click”-reaction. The purity of the synthesized polymer conjugates was proven via NMR spectroscopy, SEC, MALDI-ToF MS and LC/MALDI-ToF MS. The behavior of the polymer conjugates in lipid membranes (DPPC) was investigated a) in monolayers via compression isotherms coupled with epifluorescence microscopy and AFM and b) in bilayers using confocal microscopy. It was shown that both polymer conjugates show similar membrane interactions, however the “2-arm structure” is better miscible with the lipids.
