Abstract: Multipotente mesenchymale Stromazellen (MSC) sind Teil der Knochenmarknische, ihre funktionelle Rolle für die gesunde Hämatopoese, ihr Einfluss bei der Pathogenese maligner Erkrankungen sowie ihr therapeutischer Nutzen sind Gegenstand aktueller Forschung. Ziel dieser Arbeit war es, durch die retrospektive Analyse eines umfangreichen Datensatzes (N=328) Parameter zu ermitteln, welche die Präsenz von MSC im Knochenmark beeinflussen und somit ihre Gewinnung erschweren. Nach Isolierung der mononukleärer Zellen aus dem Knochenmark mittels Dichtegradientenzentrifugation und Adhärenzselektion erfolgte die Identifizierung und Kultivierung der MSC standardisiert nach Dominici et al. Der Einfluss verschiedener probandenabhängiger Faktoren auf die Endpunkte MSC-Präsenz, Anzahl MSC pro ml KM sowie die Expansionsdauer wurde analysiert. Durch die analysierten Parameter kann die Auswahl der optimalen Spender-MSC anhand klar definierter Kriterien getroffen werden. Dies stellt die Grundlage zur Verbesserung der Ausgangsbedingungen bei der therapeutischen Anwendung von MSC dar. Den vorliegenden Ergebnissen nach zu schlussfolgern, scheinen MSC von jungen, gesunden Probanden für allogene Therapieansätze besonders geeignet zu sein.
Multipotent mesenchymal stromal cells (MSC) reside in the human bone marrow and may contribute to malignant disease as well as represent a tool for cellular therapies. Several factors influencing the presence of MSC have been reported, however systematic analysis of a large population of unselected donors is missing. MSC were isolated from bone marrow aspirates of 328 consecutive donors by standard methods via adherence selection and density gradient centrifugation. Presence of colony forming units-fibroblast was evaluated and cells were characterized as MSC by standard criteria. Factors influencing the success in isolation of MSC were evaluated, respectively presence, cell count per ml bone marrow and culture time. This analysis in a large unselected population identified distinct factors associated with failure of bone marrow MSC isolation. These findings could help selecting optimal MSC donors for therapeutic approaches. Based on the present study young and healthy subjects seems to be the most suited pool for high quality MSC.
