dc.description.abstractLieferung von chemischen Verbindungen: • Verbindungen wurden basierend auf ihren hemmenden Wirkungen auf posttranslationale Modifikation en gemäß früheren Studien ausgewählt • Vorab Screening von Verbindungen auf ihre hemmende Wirkung auf das Wurzelwachstum und den mitotischen Zellzyklus in Gerste (Zebularin, Trichostatin A, BIX 01294) • Verbindungen wurden mit Nadeln injiziert, um sie in meiotische Zellen zu bringen (Ahn et al.) Einzelpollen Genotypisierung • Einzelpollenkern Genotypisierung mittels digitaler PCR ohne WGA wurde etabliert (Ahn et al. 2021). • Pflanzen mit unterschiedlicher Genom und Kerngröße sind mit der etablierten Methode kompatibel. • Unterschiede in den Rekombinationsraten wurden gemessen i) Pflanzen, die unter verschiedenen Wachstumsbedingungen angebaut warden ii) verschiedene Bestocker einzelner Pflanzen iii) chemisch behandelte vs. unbehandelte Pflanzen • Einige der Zebularin injizierten Pflanzen zeigten eine zweifach e Erhöhung der Rekombinationsrate des zentromerischen Intervalls.ger

Single pollen genotyping: • Single pollen nucleus genotyping via digital PCR without WGA was established (Ahn et al . 2021) • Plants with different genome & nucleus size are compatible with the established method • Differences in the recombination rates were measured for i) plants grown under different growth conditions ii) different tillers of single plants iii) chemical treated vs. untreated plants • Some of the Zebularine injected plants showed a two fold increase in the recombination rate of the centromeric intervaleng
