dc.description.abstractIn dieser Arbeit wurden die Mutantenlinien Glucosinolate Content and Composition 7 (gcc7) und gcc8 funktionell analysiert. Die Mutation in gcc8 führt zum Funktionsverlust des Schwefeltransporters SULTR1;2 und zu verringertem Glukosinolat- (GSL-) gehalt. SULTR1;2 beeinflusst den primären und sekundären Schwefelhaushalt. Durch Einzelnukleotidaustausche wurden Varianten generiert, um Transport- und Signalfunktion von SULTR1;2 als potentiellen Transzeptors getrennt zu untersuchen. In gcc7 weist IQD1 eine erhöhte Expression auf. IQD-Proteine binden Calmodulin und spielen wahrscheinlich eine Rolle in der Ca2+ Signalkette. IQD1 ist an Mikrotubuli und im Zellkern lokalisiert. In dieser Arbeit wurde erstmals die Funktion von IQD1 im Zellkern und für die Genexpression untersucht. Die neu generierten IQD1-Missexpressionslinien wiesen keinen veränderten GSL- oder Phytohormongehalt auf. Eine mögliche Funktion in der Pflanzenabwehr und GSL-Biosynthese von IQD1 zusammen mit KLCR2, CAM2 und WRKY21 wurde in Transaktivierungsexperimenten untersucht.ger
dc.description.abstractIn this study mutants Glucosinolate Content and Composition 7 (gcc7) and gcc8 are functionally characterized. The mutation in gcc8 leads to a loss of function of the major sulfate transporter SULTR1;2 and to lower glucosinolate (GSL) content. SULTR1;2 affects the primary and secondary sulfur metabolism. Variants were generated via single nucleotide exchanges to uncouple transport and signaling function of the potential Transceptor SULTR1;2. In gcc7 the gene locus of IQD1 shows elevated expression. IQDs are CaM binding plant proteins and might play a role in Ca2+ signaling cascades. IQD1 is associated with microtubules (MT) and localized to the nucleus. For the first time this work focuses on the nuclear IQD1 function in gene expression. Newly generated IQD1 missexpression lines showed no altered GSL or phytohormone content compared to the wild type. A function in plant defense and GSL biosynthesis of IQD1 in in a complex together with KLCR2, CAM2 and WRKY21 were analysed via a transactivation assay.eng
