Abstract: Die CXXC-Oxidoreduktasen steuern eine Vielzahl zellulärer Redoxprozesse. Dabei ist unklar, wie deren Proteinstruktur die beobachtbare Aktivitätsbreite erzeugt. Ziel der Arbeit war es durch den Transfer des CXXC-Tetrapeptidmotivs in das artifizielle Protein M7 neue Erkenntnisse über die Struktur-Funktionsbeziehung dieser Oxidoreduktasen zu erhalten. Die Implementierung der active site Cys-X-X-Cys-Sequenz, aus Thioredoxin und DsbA, erzeugte im M7 ein nukleophiles Cystein dessen Aktivität mit der M7-Struktur gekoppelt ist. Dabei zeigten M7-Redoxvarianten in thermodynamischen (Redoxpotential und Cystein pKa) und kinetischen Assays (Redoxkinetik) signifikante Unterschiede zu den Kontroll-CXXC-Peptiden. Hierbei ist die Redoxaktivität mit der M7-Struktur komplexer verknüpft als in den natürlichen Oxidoreduktasen. Diese Arbeit stellt den initialen Designzyklus zur Generierung von redoxaktiven M7-Varianten dar.
The CXXC-oxidoreductases perform a range of cellular redox processes. However it is still unknown, how these enzymes generate their huge functional diversity, despite minimal structural differences. The transfer of the CXXC sequence into the artificial protein M7 can illuminate the relationship between structure and function of these oxidoreductases. Aim of this work was the design of a redox active M7. The implementation of the thioredoxin and DsbA tetrapeptid motif (Cys-X-X-Cys) into M7 generated a structure dependent nucleophilic cysteine. In thermodynamic (cysteine E° and pKa) and kinetic assays (redox kinetic) M7 redox variants showed significant activities compared to control CXXC-peptides. For all M7 redox variants, the redox activity is stronger convoluted with the structure than in natural oxidoreductases. Here the stability of the native oxidized state appears as the key regulator. This work represents the successful initial design round to construct redox active M7 variants.
