dc.description.abstractEs wurde nachgewiesen, dass nicht nur der lösliche Ligand des Epidermalen Wachstumsfaktors (EGF), sondern auch seine Proform (proEGF) funktionell aktiv ist. Es konnte gezeigt werden, dass die Expression der cytoplasmatischen Domäne des EGF (proEGFcyt) sowohl die Proliferation, Motilität als auch Migrationsfähigkeit der humanen follikulären Schilddrüsenkarzinomzelllinie FTC-133 hemmt. Als potentiell interaktive Domäne wurde dabei die von Exon 23 kodierte Sequenz auf proEGF ermittelt. Die Expression der in dieser Arbeit neu identifizierten proEGF-Isoform (proEGFdel23) konnte diese Effekte nicht induzieren. Sowohl mRNA-Expressionsstudien als auch funktionelle Untersuchungen indizieren eine tumorregressive Wirkung von proEGFcyt in humanen Schilddrüsenkarzinomzellen. ProEGFcyt greift in die Regulation cytoskelletaler Funktionen ein, wobei die Induktion der Produktion des Mikrotubuli-assoziierten Proteins MAP2c einen potentiellen Schlüsselfaktor in der proEGFcyt-vermittelten Proliferations- und Motilitätshemmung bildet. Die MAP2c-vermittelte Inhibition der Mikrotubulidynamik sowie der Mikrotubuli-Kinesin-Interaktion stellen hier potentiell limitierende Faktoren in den Prozessen von Proliferation, Motilität und Migrationsfähigkeit dar. Durch das komplexe Interagieren von Cytoskellet und vesikulärem Kompartment alteriert proEGFcyt zudem die Abläufe im Vesikeltransport von FTC-133 und inhibiert damit potentiell die Migrationsfähigkeit der Tumorzelle durch extazelluläre Matrixproteine.-
dc.description.abstractIt could be demonstrated that not only the soluble hormone of Epidermal Growth Factor (EGF) but also its precursor hormon is a functional active molecule. It could be shown that the expression of cytoplasmic domain of EGF (proEGFcyt) is inhibiting as well proliferation, motility as migration of the human follicular thyroid carcinoma cell line FTC-133. The region coded by exon 23 of proEGF could be identified as the potentially interactive domain of proEGFcyt. The in this dissertation new identified proEGF-Isoform (proEGFdel23) was not able to induce the same effects as proEGFcyt. As well mRNA expression studies as functional investigations indicate an anticancerous effect of proEGFcyt in human thyroid carcinoma cells. ProEGFcyt is interfering in the regulation of cytoskeleton functions. Here induction of production of the Microtubule-associated Protein MAP2c offers a potential key factor in proEGFcyt-induced inhibition of proliferation and motility. MAP2c-mediated inhibition of microtubule dynamics as well as inhibition of microtubule-Kinesin-interactions offer potential limiting factors in the processes of proliferation, motility and migration. Upon the complex interactions of cytoskeleton with vesicular compartment proEGFcyt also seems to alter vesicle transport of FTC-133 and therefore also potentially mediates inhibition of migration of the tumour cell through extracellular matrix proteins.eng
