Abstract: Die Alkaloide des Benzylisochinolintyps kommen in ungefähr 2500 Strukturen vor. Zu ihnen gehören viele physiologisch aktive Substanzen. Innerhalb dieser Gruppe von Alkaloiden ist Morphin der pharmazeutisch wichtigste Vertreter. Es wird immer noch aus dem Schlafmohn Papaver somniferum gewonnen. Schlafmohn produziert, ausgehend vom L-Tyrosin, mehr als 80 verschiedene Alkaloide. Ein Schritt innerhalb der Biosynthese vom L-Tyrosin zum Morphin ist die Azetylierung des Salutaridinols durch die Salutaridin-7-O-Azetyltransferase (SalAT) mit anschließender Deazetylierung unter Bildung des pentazyklischen Morphinan Ringsystems des Thebains. SalAT wurde zuerst, ausgehend von einer Aminosäureteilsequenz des aus Zellsuspensionskulturen von Papaver somniferum, gereinigten nativen Proteins, als Volllänge cDNA Klon isoliert (Grothe et al., 2001). SalAT katalysiert die Umsetzung des Phenanthrenalkaloides Salutaridinol zum Salutaridinolazetat, die direkte Vorstufe des Thebains innerhalb der Morphinbiosynthese. Da Salutaridinolazetat schnell nicht-enzymatisch zerfällt, konnte bisher nicht geklärt werden, ob die Thebainbildung enzymatisch oder nicht-enzymatisch erfolgt. Es gab einige Hinweise darauf, das die Eliminierung ein enzymatischer Prozess ist. Wegen Instabilität des Azetates wurde die "Thebainsynthase" als physisch mit der Salutaridin-7-O-Acetyltransferase verbunden aufgefasst. Aufbauend auf dieser Hypothese wurde ein Yeast-two-hybrid System zur Isolierung einer für die putative Thebainsynthase kodierenden cDNA genutzt. Yeast-two-hybrid Assays zur Isolierung von SalAT interagierenden Proteinen ergaben eine relativ hohe Anzahl von cDNAs als Kandidaten für die Thebainsynthase. Basierend auf unserer biologischen Datenanalyse schlussfolgern wir, dass die Thebainbildung im Schlafmohn eher ein nicht-enzymatischer denn ezymatischer Schritt ist, höchst wahrscheinlich wird sie vom Major Latex Protein (MLP146) beeinflusst.
Benzylisoquinoline alkaloids represent a group of about 2500 structures containing many physiologically active members. Among the benzylisoquinoline alkaloids, morphine is one of the pharmaceutically important members that are still derived from the opium poppy Papaver somniferum, which produces over 80 alkaloids derived from L-tyrosine. Through the biosynthetic pathway that leads from L-tyrosine to morphine, salutaridinol is acetylated by salutaridinol 7-O-acetyltransferase (SalAT) and, subsequently, the acetate group is removed to form the pentacyclic morphinan ring system of thebaine. SalAT was initially identified and isolated as a cDNA clone that corresponds to the internal amino acid sequences of the native enzyme purified from a cell suspension culture of Papaver somniferum (Grothe et al., 2001). SalAT catalyzes the conversion of the phenanthrene alkaloid salutaridinol to salutaridinol acetate the immediate precursor of thebaine along morphine biosynthetic pathway. Because salutaridinol acetate non-enzymatically rapidly decomposes to thebaine, it was not clarified whether in vivo thebaine formation is an enzymatic or a non-enzymatic reaction. There were some indications which suggest that this elimination is an enzymatic - dependent process. Because of the instability of the acetate, this "thebaine synthase" was assumed to be physically correlated with salutaridinol 7-O-acetyltransferase. Based upon this hypothesis, a yeast two-hybrid system was used in an attempt to isolate a cDNA that encodes a putative "thebaine synthase". Yeast two-hybrid assays for isolation of SalAT interaction proteins; pulled out a relatively high number of cDNA as candidates for "thebaine synthase". Based on our the biochemical data analysis we suggest that in vivo thebaine formation in opium poppy is a non enzymatic rather than enzymatic step, most likely influenced by major latex protein (MLP 146).
