Abstract: Das druckempfindliche Modellprotein VHP35 und die Variante VHP35_L69A wurden für eine umfassende biophysikalische Analyse unter wechselnden physikalischen Bedingungen mittels Lösungs-NMR untersucht. Innerhalb des experimentell zugänglichen Temperatur- und Druckbereichs weist VHP35 eine hyperbolische Phasengrenze im Temperatur-Druck-Phasenraum auf, die das Ergebnis von druckbedingten stabilisierenden volumetrischen Umlagerungen ist die zu einer höheren Kompressibilität im nativen Zustand im Vergleich zum entfalteten Zustand führt. Der letztgenannte Unterschied in der Kompressibilität verschwindet durch die L69A-Substitution, was zu einer elliptischen Phasengrenze und einer entropischen, druckinduzierten Destabilisierung führt. Bei 278 K zeigen sich strukturell deutliche Konformationsunterschiede für die druckabhängigen Konformeren zwischen dem Wildtyp und der Variante, im Wesentlichen innerhalb des Rückgrats und insbesondere in der C-terminalen Helix.
The pressure-sensitive model protein VHP35 and the variant VHP35_L69A were studied for comprehensive biophysical analysis under changing physical conditions by solution NMR. Within the experimentally accessible temperature and pressure range, VHP35 reveals a hyperbolic phase boundary in the temperature-pressure phase space, which is the result of pressure-induced stabilizing volumetric rearrangements leading to higher compressibility in the native state compared to the unfolded state. The latter difference in compressibility disappears by the L69A substitution resulting in an elliptic phase boundary and entropic pressure-induced destabilization. At 278 K, structurally distinct conformational differences for the pressure-dependent conformers are revealed between the wild type and the variant, essentially within the backbone and particularly in the C-terminal helix.
