dc.description.abstractJasmonate sind ubiquitär verbreitete Signalstoffe in Pflanzen, die sowohl an der Vermittlung von Stressantworten als auch an der Regulation von Wachstum und Entwicklung beteiligt sind. Die Allenoxidcyclase (AOC) ist ein Schlüsselenzym der Biosynthese der Jasmonsäure (JA), da sie die Bildung der cis(+)-12-Oxophytodiensäure (OPDA) katalysiert und damit für die Etablierung der enantiomeren Grundstruktur der natürlich vorkommenden Jasmonate verantwortlich ist. In Arabidopsis thaliana kodieren vier Gene für AOCs. Ziel dieser Arbeit war die funktionale Charakterisierung der AOC-Genfamilie aus A. thaliana, insbesondere die Frage nach der spezifischen oder redundanten Funktion einzelner AOCs. Durch transgene loss of function-Ansätze, Promotor-GUS-Analysen sowie Expressionsstudien konnte gezeigt werden, dass von einer partiellen funktionalen Redundanz einzelner AOCs in unterschiedlichen Prozessen ausgegangen werden muss. Ein weiteres Ziel der Arbeit bestand in der Analyse der β-Oxidationsschritte der JA-Biosynthese. Anhand von Mutanten mit Störungen in der Peroxisombiogenese (pex6) bzw. der peroxisomalen Fettsäure-ß-Oxidation (aim1) konnte die Notwendigkeit funktionaler Peroxisomen für die Biosynthese von Jasmonaten bestätigt werden. Weiterhin konnte erstmals die Beteiligung des multifunctional proteins AIM1 an der wundinduzierten JA-Bildung nachgewiesen werden.-
dc.description.abstractJasmonates are ubiquitously occurring signalling compounds involved in plant stress responses and the regulation of plant growth and development. A key enzyme in the biosynthesis of jasmonic acid (JA) is the allene oxide cyclase (AOC) which is responsible for the exclusive formation of the correct stereoisomeric precursor, cis(+)-12-oxophytodienoic acid (OPDA). In Arabidopsis thaliana four genes coding for functional AOCs were identified. The primary aim of this work was the functional characterization of the AOC gene family with a focus on the question of specific or redundant functions of the individual AOCs. Transgenic loss of function approaches, analysis of promoter-GUS-lines as well as expression studies indicated that different AOCs function partially redundant in differential processes. Another aim of this work was the analysis of the β-oxidation steps required for the formation of JA. The analysis of Arabidopsis mutants which are affected in peroxisome biogenesis (pex6) or disrupted in fatty acid ß-oxidation (aim1) confirmed the necessity of functional peroxisomes for JA-Biosynthesis. Furthermore, the involvement of the multifunctional protein AIM1 in wound-induced JA-formation was shown for the first time.eng
