Abstract: Angiogeneseinhibitoren spielen in der modernen Tumortherapie in Kombination mit herkömmlichen Chemotherapeutika eine wesentliche Rolle. Ihre Wirkung stützt sich auf die These, dass der wachsende Tumor ab einer bestimmten Größe über Diffusion nicht mehr ausreichend mit Nährstoffen und Sauerstoff versorgt werden kann. Für ein weiteres Wachstum ist eine eigene, durch das entartete Gewebe induzierte Neoangiogenese erforderlich. Angiogeneseinhibitoren sollen diesen Prozess stoppen und damit zur Tumorregression führen. Bisher widersprüchlich diskutiert wurde die Frage, ob eine Kombination von Angiogenesehemmung und Bestrahlung synergistische Effekte erbringt. Zur Untersuchung der Fragestellung wurde ein Tumormodell auf der Chorionallantoismembran (CAM) des bebrüteten Hühnereies entwickelt, an dem sowohl Versuche zur Angiogenesehemmung, Bestrahlung als auch Kombination beider Therapieformen durchgeführt werden konnten. Die Auswertung der Ergebnisse erfolgte nach histologischer Aufarbeitung mit verschiedenen Färbemethoden. Mit dieser Arbeit konnte gezeigt werden, dass sich das Chorionallantoismembran-Modell zur Untersuchung tumorbiologischer Fragestellungen im besonderen eignet. Für die Kombination von Bestrahlung und Angiogenesehemmung konnten synergistische Effekte in Form von Tumorregression, Verringerung der Gefäßanzahl sowie des VEGF-Rezeptor 2-Besatzes beim Rhabdomyosarkom nachgewiesen werden. Die beobachteten Effekte lassen eine Kombination von Angiogenesehemmung und Bestrahlung als viel versprechenden Therapieansatz erscheinen.
Inhibitors of angiogenesis play a pivotal role in the treatment of malignant tumours in combination with conventional chemotherapeutic agents. When the tumour grows up to a size of 2-3 mm3 the supply with nutrients and oxygen becomes inadequate. Growth of a tumour beyond this size requires development of a microvessel network. The tumour induces a neoangiogenesis. Antiangiogenetic agents should stop this process and induce tumour regression. However, conflicting reports exist for a combination of radiation and antiangiogenetic therapies. To study these effects we established a tumour model on the chorioallantoic-membrane (CAM) of the chick embryo. The model was used to investigate the antiangiogenic effects, radiation effects and a combination therapy. Results were analysed using histological staining. It could be shown that the CAM is suitable for studying the biology of tumour angiogenesis. The combination of radiation and antiangiogenetic agents show synergistic effects in respect of tumour regression, reduction of tumour vessels and VEGF-Receptor-2. These findings suggest that a combination of radiation and inhibition of angiogenesis results in a promising treatment in the tumour therapy.
