Abstract: In der vorliegenden Arbeit wird die säulenchromatographische Reinigung einer Peptidyl-Prolyl-cis/trans-Isomerase (PPIase) aus Pflanzen, speziell aus Proembryogenen Massen (PEMs) von Digitalis lanata beschrieben. Diese, zur Familie der phosphospezifischen Parvuline gehörende PPIase bevorzugt Peptidsubstrate, die das Motiv pThr/pSer-Pro aufweisen. Weiterhin wird die Klonierung, die heterologe Expression und die biochemische Charakterisierung von DLPar12.8 dargestellt. Durch erste nähere Aussagen, z.B. über Substratspezifität, Inhibierbarkeit und Verteilung in der Pflanze, sollte ein Vergleich mit bereits bekannnten Vertretern der phospho-spezifischen Parvuline ermöglicht werden. Im Gegensatz zu diesen bereits bekannten Vertretern der phosphospezifischen Parvuline zeichnet sich DLPar12.8 durch das Fehlen einer Phosphopeptide bindenden WW-Domäne aus. Es konnte gezeigt werden, daß trotz des Fehlens der WW-Domäne DLPar12.8 vermutlich eine ähnliche Funktion ausübt, wie das humane Pin1 bzw. ESS1 aus Hefe.
A peptidyl-prolyl cis/trans isomerase (PPIase) was isolated from proembryogenic masses (PEMs) of Digitalis lanata according to its enzymatic activity. Partial sequence analysis of the purified enzyme (DLPar12.8) revealed sequence homology to members of the parvulin family of PPIases. Similar to human Pin1 and yeast Ess1, it exhibits catalytic activity towards substrates containing pThr/pSer-Pro peptide bonds and comparable inhibition kinetics with juglone. Unlike Pin1-type enzymes it lacks the phosphopeptide binding WW-domain. Western blotting with anti-DLPar12.8 serum recognized the endogenous form in the cytosolic fraction of the plant cells. Since the PIN1 homologue ESS1 is an essential gene, complementation experiments in yeast were peformed. It reveals that DLPar12.8 is almost as effective as the nuclear hPin1 in rescuing the temperature sensitive phenotype of the Saccharomyces cerevisiae strain, which is caused by a mutation in ESS1. In contrast, the human parvulin hPar14 is not able to rescue the lethal phenotype of this yeast strain at non-permissive temperatures. These results suggest a function for DLPar12.8 rather similar to parvulins of the Pin1-type.
