Abstract: Im Rahmen der vorliegenden Arbeit wurde ein tumorspezifisches Vektorsystem entwickelt. Dieses Vektorsystem ist modular aus virusanalogen Partikeln (VLP's) des Polyomahüllproteins VP1 und Fv-Fragmenten des tumorspezifischen Antikörpers B3 aufgebaut. B3 ist gegen das Antigen Lewis Y gerichtet, das auf vielen humanen Tumoren präsentiert wird. Damit verschiedene funktionelle Module, wie Targeting-Domänen, auf der Oberfläche von Polyoma-VLP's präsentiert werden könnnen, wurde eine Methode zur spezifischen Kopplung von Biomolekülen entwickelt. Zwei komplementär geladene Peptide, die jeweils ein Cystein enthalten, reagieren in Gegenwart eines Redoxsystems zu einem chimären Polypeptid, einem Heterodimer. Dabei werden die geladenen Peptide mittels polyionischer Wechselwirkungen spezifisch assoziiert und anschließend durch eine Disulfidbrücke zwischen den Cysteinen kovalent verknüpft. Um diese effektive Kopplungsmethode auf das modular konstruierte Vektorsystem zu übertragen, wurde das Antikörperfragment und das Hüllprotein VP1 mit diesen polyionischen Peptiden fusioniert. Zum einen wurde der C-Terminus der VH-Domäne um die Peptidsequenz Arg8CysPro verlängert, zum anderen das komplementär geladene Peptid Glu8Cys in den lösungsmittelexponierten HI-Loop von VP1 (VP1-E8C) inseriert. Die VP1-Variante VP1-E8C wurde rekombinant in E. coli produziert und zum homogenen pentameren Protein gereinigt. In Gegenwart von Ammoniumsulfat assemblierten diese VP1- Pentamere ohne Kontamination durch infektiöse virale RNA oder DNA zu VLP´s. Die in vitro rekonstituierten Partikel wurden mittels Gelfiltration, analytischer Ultrazentrifugation und Elektronenmikroskopie als globuläre, kapsidähnliche Strukturen nachgewiesen. Durch die gezielte Mutagenese des HI-Loops wurde nicht nur der Adapter für die Kopplung des Antikörperfragments eingeführt, sondern auch die Sialinsäure-Bindungsstelle der Polyoma-VLP´s blockiert, die sich unterhalb des HI-Loops befindet. Dadurch gelang es, die natürliche Zellbindung der VLP´s zu inaktivieren. Durch die Kopplung des B3-Fv-Fragments an die VLP´s konnte dem Vektorsystem somit eine Zelltypspezifität verliehen werden. Auf der Oberfläche eines VLP´s von VP1-E8C wurden ca. 30 Antikörperfragmente präsentiert. Mittels dieses modular-aufgebauten Transportsystems wurde DNA verpackt und Säugerzellen transfiziert. Aufgrund der erworbenen Zelltypspezifität der VLP´s wurden nur Zellen transfiziert, die das tumorspezifische Antigen Lewis Y auf der Zelloberfläche präsentieren.
In this thesis a tumorspecific gene-transfer system was constructed. This delivery system is characterized by its modular structure based on virus like particles (VLP´s) of the polyomavirus major coat protein VP1 and the Fv-fragments of the tumorspecific antibody B3. B3 recognies the antigene Lewis Y, present on several human tumor cells. To allow the presentation of different functional moduls on the surface of Polyoma VLP´s a method for specific coupling of biomolecules was developed. In presence of a redox system two complementary charged peptides, each containing one cysteine residue, react to a chimeric polypeptide - a heterodimer. First the charged peptids specifically associated via polyionic interactions, and then they are covalently linked by a disulfide bond. In order to use this efficient conjugation method for the modular-designed vector system the antibody fragment and the coat protein VP1 was fused with polyionic peptides. On the one hand the C-terminus of the VH-domain was extended by the peptide sequence Arg8CysPro and on the other hand the complementary charged peptide Glu8Cys was inserted in the solvent exposed HI-loop of VP1 (VP1-E8C). The mutant VP1-E8C was recombinantly produced in E. coli and purified to homogenous pentameric protein. In presence of ammonium sulphate these VP1-pentamers assembled into VLP´s without any infectious viral RNA or DNA. The in vitro reconstructed particles were analyzed by size exclusion chromatography, analytical ultracentrifugation and electron microscopy. The inserted peptide in the HI-loop served not only as adapter for coupling of antibody fragments, but also inhibited the sialic acid binding site of Polyoma VLP´s. Thus the native cell attachment ability was abolished. The celltyp-specific retargeting of the particles were obtained by coupling B3-Fv-fragments to VLP´s. Approximately 30 antibody fragments were presented on the surface of one VP1-E8C-VLP. Using this modular-designed delivery system DNA was packaged and mammalian cells transfected. Caused by the celltype specificity of VLP´s cells were only transfected presenting the tumorspecific antigene Lewis Y.
