Abstract: Ziel der Arbeit war es, die Aussagefähigkeit der Speicheltestung auf zyto- und gentoxische Effekte zu überprüfen. Ein deutlicher Zusammenhang zwischen zytotoxischer Wirkung des Speichels und dem Rauchverhalten der Probanden konnte mit der Bestimmung der 'plating efficincy' bei Säugerzellen gezeigt werden. Ein vergleichbarer Zusammenhang lag auch zwischen der Gentoxizität der Proben und dem Raucherstatus vor. Gentoxische Unterschiede im Speichel von Rauchern und Nichtrauchern konnten allerdings erst statistisch abgesichert werden, wenn auch der Alkoholkonsum berücksichtigt wurde. Ein modifizierender Faktor könnte sein, dass aufgrund der relativ großen Personenzahl auf eine vorgegebene ,kontrollierte nichtmutagene Ernährung verzichtet wurde. Allerdings kamen unsere Probanden aus einem vergleichsweise engen Kulturkreis mit ähnliche Ernährungsformen und daher wurde dieser Einflussfaktor vernachlässigt. Das Alter hingegen stellte keinen modifizierenden Faktor dar. Es ist also zu erkennen, dass die zyto- und gentoxische Wirkung des Speichels ein Anzeichen dafür ist, dass die Mundschleimhaut verstärkt belastet und einem "kanzerogenen Stress" ausgesetzt ist. Damit wiederum sind die Möglichkeiten eines Präscreeningprogramms von Risikogruppen im Rahmen der Früherkennung von Tumoren im Mund- und Rachenraum gegeben.
Target of the work was it to check the informative capability of the saliva testing for cytotxic and gene-toxic effects. A clear connection between cytotoxic effect of the saliva and the smoke behaviour of the pro gangs could be shown with the regulation 'plating efficiency' with mammalian cells. A comparable connection was present also between the gene toxicity of the samples and the smoker status. Gene-toxic differences in the saliva of smokers and non-smokers could be secured however only statistically even if the consumption of alcohol were considered. A modifying factor could be that due to the relatively large number of persons without a given, controlled non-mutagene nutrition one did. However our pro gangs came from a comparatively close culture area with similar nutrition forms and therefore this factor of influence were neglected. The age however did not represent a modifying factor. It is to be detected thus that zyto and towards-toxic effect of the saliva is a sign for it that the mouth mucous membrane is exposed strengthened loaded and to a "carcinogenic stress". Thus again the possibilities of a praescreeningprogramm of groups of risks are given in the framework the early recognition of tumours in the mouth and throat space.
