dc.description.abstractIn der pharmazeutischen Industrie steigt die Bedeutung der topischen Formulierungen stetig an. Um deren Verwendungen zu erweitern, müssen die Formulierungen ständig weiter optimiert werden. Die Forderungen an topischen Formulierungen sind sehr unterschiedlich. Für jedes gewünschte Konzentration-Zeit-Profil kann die Zusammensetzung aus einer Anzahl üblicher Rezepturen ausgewählt werden. Weiterhin kann das Penetrationsverhalten durch Zugabe von Bestandteilen mit typischen Eigenschaften beeinflusst werden. Da der Penetrationsprozess durch die Haut sehr komplex und von vielen Faktoren abhängig ist, kann der Einfluss der Formulierungskomponente auf den Penetrationsprozess nur mit Hilfe eines mathematischen Modells geklärt werden. In der vorliegenden Arbeit wurde der Freisetzungs- und Penetrationsprozess aus einer Suspension durch ein mathematisch-physikalisches Modell beschrieben. Als Formulierungsgrundlage wurde Vaseline verwendet. Im Modell wurden die Einflüsse der einzelnen Suspensionskomponente charakterisiert. Durch eine Änderung des Anteils an dünnflüssigem Paraffinanteil in der Suspension wurde das Freisetzungs- und Diffusionsverhalten in der Suspension beeinflusst. Mit Hilfe des mathematischen Modells wurde diese Abhängigkeit quantitativ charakterisiert. Im untersuchten System wurde eine Dodecanol-Collodium-Membran als Akzeptor verwendet. Das Penetrationsverhalten vom Wirkstoff im Akzeptor wurde durch ein Zugabe von Propylenglykol in die Suspension beeinflusst. Hier fungiert Propylenglykol als Enhancer. Mit Hilfe des Modells wurde die quantitative Abhängigkeit zwischen dem Enhancer-Effekt und der Enhancer-Konzentration in der Suspension charakterisiert. Mit dem Modell wurden die Simulation der Freisetzung und der Penetration aus Suspensionen mit unterschiedlicher Zusammensetzung unter in-vivo Bedingungen durchgeführt. Die Simulationsergebnisse erlauben eine Vorhersage über die Formulierungszusammensetzung mit optimalem Freisetzungs- bzw. Penetrationsverhalten. Da ein Optimierungsverfahren mit Hilfe eines mathematischen Modells jedoch höherere Forderungen an Parametern als die bisherigen Optimierungsverfahren benötigt, sind die gängigen Untersuchungsmethoden wie z.B. Franz Zelle oder das Mehrschichtmembransystem (MSMS) für die vorliegende Arbeit nicht geeignet. Diese Methoden können die benötigten Parameter nur mit sehr großem Aufwand bestimmt werden. Die Genauigkeit der Methoden ist zudem für das mathematische Modell nicht ausreichend. Aus diesem Grund wurden in der Arbeit die spektroskopischen Methoden, die abgeschwächte Totalreflexion (ATR) und die photoakustische Spektroskopie (PAS), für die Penetrationsuntersuchung angepasst.-
dc.description.abstractNowadays, there is a growing interest for optimising topical formulations. The demands on topical formulations are very different. For any concentration-time profile desired, the composition can be selected from a number of usual recipes. Adding of ingredient of specific properties can influence the penetration behaviour. Because the drug delivery through the human skin is very complex and depends on many factors, using a mathematical model can only elucidate the influence of various components of the formulation on the penetration process. The present work is devoted to describe the release and penetration of a drug from a suspension based on a mathematical-physical model. Vaseline is used as basic component of the formulation. The influence of each particular component of the suspension was characterised. Changing the amount of paraffinum liquidum in the suspension the release and penetration behaviour were affected. Based on the appropriate mathematical model the relevant dependencies were quantitatively characterised. An artificial dodecanol-collodion membrane was used as acceptor. Propylene glycol was used as enhancer. The relationship between enhancer effect and enhancer concentration in the suspension is quantitatively described. The drug release and penetration from a suspension were simulated under in vivo conditions. The results of the simulation facilitate formulation design so that an optimal system could be developed. Because of the complexity, the mathematical model demands many parameters with high accuracy as input data, which cannot be obtained by using traditional methods like Franz's cell or multilayer membrane system. For that reason, non-invasive spectroscopic methods were applied to penetration studies, namely Fourier transform infrared attenuated total reflection (FTIR-ATR) and FTIR photoacoustic spectroscopy.eng
