Abstract: Zwergrost (Puccinia hordei) verursacht weltweit hohe Ertragseinbußen in der Gerstenproduktion, wodurch die Identifizierung neuer und wirksamer Quellen für Resistenzgene von großem Interesse ist. Eine hochauflösende Kartierungspopulation von 537 RILs mit einer Auflösung von 0.01 % Rekombination wurde konstruiert. Anhand verschiedener molekularer Marker wurden 56 Marker für die Markerabsättigung entwickelt. Schließlich wurde das Zielintervall auf 0,05 cM verkleinert, wobei elf Marker nachgewiesen wurden, die mit RphMBR1012 co-segregieren. Acht HC- und 15 LC-Gene, inklusive fünf Krankheitsresistenzgene, wurden in der Zielregion von 0.35 Mb nachgewiesen. Die Ergebnisse des diagnostischen Tests zeigten, dass sich die Marker GBS546 und GBS626 am besten für die markergestützte Selektion von RphMBR1012 eignen. Die allelspezifische Re-Sequenzierung der Gene ergab eine Größe von 17.107 bp im MBR1012 und 16.963 bp im Scarlett. Färbungen zeigten, dass die Resistenz gegen Zwergrost höchstwahrscheinlich auf die von RphMBR1012 kontrollierte hypersensitive Reaktion zurückzuführen ist.
The barley leaf rust (Puccinia hordei) disease seriously threatens barley production worldwide. Identification of effective sources of resistance genes against the Puccinia hordei in barley is of great interest. A high resolution mapping population of 537 RILs was constructed corresponding to a resolution of 0.01% recombination. Different molecular marker sources were implemented and 56 markers were developed for marker saturation. Finally, the target interval was shortened to 0.05 cM. Eleven markers were co-segregated with RphMBR1012. Eight low-confidence and 15 high-confidence genes were defined in target region comprising 0.35 Mb including five disease resistance genes. The results of diagnostic test showed that two markers GBS546 and GBS626 are considered as the best diagnostic markers for marker-assisted-selection of RphMBR1012. Allele specific re-sequencing of high and low confidence genes located in target interval resulted in 17,107 bp in MBR1012 and 16,963 bp in Scarlett. Utilization of different staining showed that resistance to the barley leaf rust in resistant lines is most likely due to the HR controlled by RphMBR1012.
