Abstract: Das obligat aerob lebende Bakterium Xanthomonas campestris pv. vesicatoria (Xcv) ist der Erreger der bakteriellen Fleckenkrankheit bei Paprika und Tomate. Während der interzellulären Vermehrung im Wirt, ist neben dem essentiellen T3SS auch die effektive Nutzung von verfügbaren Substraten und die Sensierung von Änderung des ROS-, Eisen und O2-Gehalts wichtig für eine effektive Kolonisierung. Aconitasen könnten aufgrund ihres labilen [4Fe-4S]-Cluster eine wichtige Rolle bei der Sensierung derartiger Bedingungen spielen und aufgrund ihrer Bifunktionalität genregulatorische Funktionen ausüben. Xcv besitzt die 3 Aconitasen AcnB, AcnA und AcnA2. Eine Deletion von acnB -und acnA2- führt zu reduziertem Wachstum in planta. Eine Deletion von acnB führt zu reduziertem Wachstum mit Citrat als C-Quelle, während eine Deletion von acnA2 das Wachstum unter diesen Bedingungen verbessert. Expressionsstudien zeigten, dass AcnA2 neben der Regulation der Expression von acnA und acnB auch in die Regulation der Expression wichtiger Gene des Kohlenhydrat- und Citratstoffwechsels, der Eisenhomöostase und der Abwehr von ROS involviert ist.
The obligate aerobic living bacterium Xanthomonas campestris pv. vesicatoria (Xcv) is the causal agent of the bacterial spot disease on pepper and tomato plants. During intercellular growth of Xcv in the host, the effective colonization relies upon the essential T3SS and the ability to adapt to changing substrate availability, as well as being able to sense and cope with changes in iron- and oxygen-levels along with ROS. Aconitases might be important proteins in adaption to such an environment because of their labile [4Fe-4S]-cluster and as well might have a regulatory role in genexpression because of their known bifunctional character. Xcv possesses the 3 Aconitases AcnB, AcnA and AcnA2. An acnB and an acnA2 mutation cause reduced growth of Xcv in planta. A deletion of the acnB gene leads to reduced growth in vitro with citrate as the carbon source, while an acnA2 mutation causes an enhanced growth. Studies of genexpression identified AcnA2 as a regulator of expression of acnB and acnA as well AcnA2 appears to be involved in regulation of genes of the citrate and carbohydrate metabolism, iron homeostatis and in protection against oxidative stress.
