Abstract: Lysin Histon Demethylasen spielen eine wichtige Rolle bei der Regulierung des epigenetischen Codes und damit in der Entwicklung von verschiedenen Krankheiten wie Krebs und neurodegenerativen Erkrankungen. Lysin spezifische Demethylase 1 (LSD1/KDM1) hat einen starken Einfluss auf die Androgen- und Östrogen-abhängige Genexpression, so dass es als ein vielversprechendes Target für die Therapie von hormonabhängigen Krebs dient. Ausgehend von vorläufigen Aktivitätsdaten von niedrigmolekularen LSD1-Inhibitoren wurden mehrere computergestützte Modelle (Structure-Based and Ligand-Based) erstellt, die in der Lage sind: 1. die Bindungsmodi von niedrigmolekularen Liganden in LSD1 zu postulieren, 2. neue Leitstrukturen an LSD1 zu entdecken. Die Arbeit bestätigt, dass computer-basiertes Wirkstoffdesign (CAMD) als ein potentes Werkzeug zur Identifizierung neuer Wirkstoffe dient.
Lysine demethylases play an important role in regulation of the epigenetic code and thus in the development of several types of human diseases such as cancer or neurodegenerative disorders. As the lysine specific demethylase 1 (LSD1/KDM1) has been strongly connected to androgen and estrogen dependent gene expression, it serves as a promising target for the therapy of hormone dependent cancer. Starting from preliminary activity data of small LSD1 inhibitors, here are reported several computational approaches (structure-based and ligand-based) able to: 1- identify new LSD1 features concerning the binding mode of small ligands inside LSD1 active cavity and 2- discover new LSD1 active leads; confirming the potentiality of computer assistant molecular design (CAMD) in medicinal chemistry as potent tool for the identification of new active compounds.
