Abstract: Neben P. aeruginosa und obligaten Anaerobiern ist S. constellatus, ein mikroaerophiles Bakterium, im CF-Sputum enthalten. Es konnte in hohen Keimzahlen bei 39 % aller CF-Exazerbationen nachgewiesen werden. Es wurden die Wechselwirkungen zwischen P. aeruginosa, S. constellatus und anderen obligaten Anaerobiern untersucht. P. aeruginosa wurde mit vier bis zu 10-8 verdünnten S. constellatus-Stämmen inkubiert. Die Laktatproduktion der Anaerobier wurde spektrophotometrisch bestimmt. Das Wachstum von P. aeruginosa wurde in Anwesenheit von S. constellatus um eine Zehnerpotenz reduziert (2,9 ± 1,5 x 109 vs 3,0 ± 5,3 x 108 KBE/ml, p<0,005). Andere Anaerobier hatten keinen vergleichbaren Effekt auf dessen Wachstum. Winzige Mengen der Anaerobier (5 x 100 KBE/ml) können eine P. aeruginosa Lösung erfolgreich infizieren. S. constellatus produziert 22,0 ± 0,3 mmol/l Laktat. Diese Menge hat jedoch keinen Einfluss auf das Wachstum von P. aeruginosa. Im Vergleich mit den Ergebnissen der anderen obligaten Anaerobier ist S. constellatus deutlich pathogener, weshalb eine stärkere Berücksichtigung in Diagnostik und Therapie der CF berechtigt erscheint.
Along with P. aeruginosa and obligate anaerobes, S. constellatus, a microaerophilic bacterium, can be found in CF sputum. It is detected in high bacterial numbers in 39 % of CF lung exacerbation. We investigated the interaction between P. aeruginosa, S. constellatus and other obligate anaerobes. P. aeruginosa was incubated together with four S. constellatus strains diluted up to 10-8 fold. Lactate production of the anaerobes was determined spectrophotometrically. P. aeruginosa growth is reduced by one order of magnitude in the presence of S. constellatus (2.9 ± 1.5 x 109 vs 3.0 ± 5.3 x 108 cfu/ml, p<0.005). Other obligate anaerobes do not have a comparable effect on P. aeruginosa growth. Even minute amounts of anaerobes (5 x 100 cfu/ml) successfully infect a P. aeruginosa solution. S. constellatus produces 22.0 ± 0.3 mmol/L lactate, but this amount of lactate does not influence P. aeruginosa growth. Comparing the results, S. constellatus appears more pathogenic than other obligate anaerobes and deserves more consideration in CF diagnosis and therapy.
