Abstract: Im Rahmen dieser Arbeit wurde die quantitave Genetik angewendet, um wichtige Gene der Terpenbiosynthese und dessen Regulation zu identifizieren. “Nested Association Mapping” (NAM) and “Genome Wide Association Mapping” (GWAS) wurde an einer Mais-NAM-Population bestehend aus 5000 rekombinanten Inzuchtlinien (RILs) angewendet, um QTLs (Quantitative Trait Locus) der Terpenbiosynthese identifizieren zu können. Für die Terpenmerkmale Linalool, (E)-Nerolidol, (E)-ß-Farnesen, (E)-ß-Bergamoten, DMNT und TMTT konnten mehrere quantitative Loci berechnet werden. Vier der QTLs konnten bis hin zum Kandidatengen eingegrenzt werden. Insgesamt wurden 5 Herbivor-induzierte Gene für diese Terpenmerkmale lokalisiert. Davon kodieren 4 Gene für Enzyme denen eine Funktion in der Terpenbiosynthese nachgewiesen werden konnte. Zusätzlich wurde ein bHLH Transkriptionsfaktor identifiziert, der möglicherweise in der Signalkaskade einen wichtigen Kontrollpunkt einnimmt. Damit konnte gezeigt werden, daß mit dieser Kartierungsstrategie QTLs bis hin zum Kandidatengen lokalisiert werden können.
In this thesis, methods of quantitative genetics were employed to identify important genes for the production of VOCs and its regulation by herbivory. “Nested Association Mapping” (NAM) and “Genome Wide Association Mapping” (GWAS) combines the advantages of association and linkage mapping. The NAM population exists of 5000 recombinant inbred lines (RILs) by crossing 25 polymorphic inbreds with the common parent line B73. NAM was used to identify quantitative trait loci (QTLs) of biosynthesis of volatile terpenes. QTLs were calculated for the traits linalool, (E)-nerolidol, (E)-ß-bergamotene, (E)-ß-farnesene, DMNT and TMTT. The QTLs were narrowed down to find candidate genes for. Finally, five herbivore-induced genes were identified. Four of these genes coded for enzymes of terpene biosynthesis that corresponded to the trait observed by the QTL. Additionally, a bHLH transcription factor was identified which may regulate the transcription of terpene biosynthesis genes. This thesis shows the opportunity to locate QTLs and their candidate genes for different traits by NAM.
