Abstract: Während des anaeroben Wachstums verstoffwechselt E. coli ein Drittel des Substrats zu Formiat. Formiat wird generiert aus Pyruvat durch das Enzym Pyruvat-Formiat-Lyase (PflB). Die Anhäufung von Formiat in hohen Konzentrationen im Cytoplasma wird durch den Transport in das Periplasma verhindert. In Abwesenheit externer Elektronenakzeptoren wird Formiat wieder in das Cytoplasma aufgenommen und dort zu CO2 und H2 durch den Formiat-Hydrogen-Lyase Komplex (FHL-Komplex) gespalten. Das FocA Protein ist in der Lage Formiat über die Cytoplasmamembran zu transportieren. Mutanten ohne FocA exkretieren weniger Formiat ins Cytoplasma, sondern häufen es in der Zelle an. Da focA-Mutanten jedoch trotz dessen noch Formiat ins Periplasma sekretieren, muss es noch mindestens ein weiteres Formiat-Transportsystem geben. Mit Hilfe eines Formiat abhängigen fdhF´-´lacZ Reportersystems wurde die Änderung der intrazellulären Formiatkonzentration untersucht. Drei verschiedene Membranproteine wurden hinsichtlich ihrer Funktion Formiat zu transportieren analysiert. Weiterhin wurde das GrcA Protein, ein Glycyl-Radikalenzym wie auch PflB, hinsichtlich seiner Funktion im anaeroben Stoffwechsel untersucht.
During fermentative growth with glucose E. coli converts up to one third of the substrate to formate. Formate is generated through the cleavage of pyruvate by the enzyme pyruvate formate-lyase (PflB). Accumulation of high cytoplasmic levels of formate is prevented by excretion of the organic acid into the periplasmic space. In the absence of exogenous electron acceptors formate is re-imported into the cell where it is disproportionates by the formate hydrogen lyase complex (FHL) into CO2 and H2. The FocA protein is able to transport formate across the cytoplasmic membrane. Mutants that are devoid of FocA excrete less formate into the medium and accumulate higher formate levels in the cytoplasm. Notably, focA mutants retain the ability to excrete and import formate, indicating the existence of at least one further system involved in formate transport. A formate-dependent fdhF´-´lacZ transcriptional fusion was used as a reporter system to monitor changes in the intracellular formate concentration. Three different membrane proteins were determined concerning the possibility to transport formate. Moreover the GrcA protein, which is also a glycyl-radical-enzyme like PflB, was determined concerning its function in the anaerobic metabolism.
