To understand better the pathogenesis of sepsis and endocarditis we analyzed the influence of Staphylococcus aureus α-toxin, Streptococcus. pyogenes SLO and E. coli LPS on the activation status of human platelets and PMN. We only examined an aggregation by α-toxin treated plate lets by aggregometry. Addition of PMN increased this aggregation rate significantly. By western blot analysis we could show thatα-toxin as well as SLO and endogenous thrombin can induce expression of Bcl-3 protein. By use of flow cytometry we determined toxin mediated activation of platelets and PMN that by use of different activation parameters. Also addition of PMN to α-toxin treated platelets increased significant platelet activation status.
