Abstract: Der Mineralokortikoidrezeptor (MR) ist in die Salz- und Wasserhomöstase involviert und entscheidend an der Blutregulation beteiligt. Auf der anderen Seite kann er aber auch pathophysiologische Effekte auf das renokardiovaskuläre System ausüben. Sowohl der MR als auch der Glukokortikoidrezeptor (GR) agieren als Liganden-gebundene Transkriptionsfaktoren am gleichen Hormon-responsiven Element. Jedoch haben sie unterschiedliche Funktionen im Körper. Eine mögliche Ursache für die pathophysiologischen MR-Effekte und die MR-Spezifität ist die posttranskriptionelle Regulation von Genen, möglicherweise über Modulation der MicroRNA-Expression. Dafür wurde untersucht, ob sich das basale MicroRNA-Expressionsprofil in humanen aortalen Endothelzellen, durch Aktivierung des MRs und GRs durch die jeweiligen Liganden ändert. Es wurden Microarrays als Screeningtest verwendet und diese Ergebnisse mittels qPCR validiert. Dabei wurde die MIR-2278 als MR-regulierte MicroRNA identifiziert. In der Zukunft sollen Targets dieser MicroRNA untersucht werden, um weitere Einblicke in die Mechanismen der Pathophysiologie des MR zu erlangen.
The mineralocorticoid receptor (MR) is involved in the blood pressure regulation by modulating the salt and water homeostasis but it can also elicit pathophysiological effects in the renocardiovascular system. The mineralocorticoid receptor (MR) and the glucocorticoid receptor (GR) bind in their active form at the same hormone response element. Surprisingly, they have different functions in the body. A possible reason for the pathophysiological effects of the MR and the specificity of the MR could be the posttranscriptional gene regulation by microRNAs. Therefore, we investigated, if the basal microRNA expression profile in human aortic endothelial cells can modified by activated MR and GR. Firstly, we examinated in Microarray studies whether special microRNAs were regulated by activated MR or GR and validated our results with qPCR. The MIR-2278 was identified as a MR-regulated MicroRNA. In the future we plan to investigate targets of the MIR-2278 to get more insights in the mechanism of the pathophysiological effects and the specificity of the MR.
