Abstract: Neuronale Stammzellen in der Zona subventricularis produzieren zwei Haupttypen von Progenitorzellen: Granule-Zellen und Periglomeruläre Zellen. Beide migrieren bis zum Bulbus olfactorius. Um den Einfluss von Aktivität auf Migration und Positionierung auf die jeweiligen Subtypen besser zu verstehen, nutzten wir gezielte postnatale Elektroporation. Hierbei konnten wir keine Unterschiede im Bewegungsmuster feststellen. Allerdings entdeckten wir einen Anstieg der Kalziumaktivität von Granule-Zellen kurz vor Erreichen ihrer finalen Position. Als wir Zellaktivität künstlich senkten, sahen wir, dass sich das Migrationsmuster kaum veränderte. Stattdessen existiert ein Einfluss auf Überleben und Positionierung von Granule Zellen. Bei Periglomerulären Zellen blieb dieser Effekt aus. Damit bewiesen unsere Experimente einen subtypen-spezifischen Effekt von neuronaler Aktivität.
Neural stem cells regionalised in the subventricular zone produce two main subtypes of neural progenitors, granule cells and periglomerular neurons. Both migrate to olfactory bulb layers. To understand the impact of activity on migration and final positioning, we used targeted postnatal electroporation to compare the two populations. We do not observe any differences regarding the mode of migration. However, we find a very striking increase of intrinsic calcium activity in granule cell precursors when they enter their target layer. By decreasing excitability in granule cell precursors, we find that neuronal activity has little effect on migration but is required for positioning and survival in the OB. Strikingly, decreasing activity of periglomerular precursors had no impact. Altogether these findings suggest that neuronal excitability is subtype specific.
