Abstract: Pflanzen als ortsgebundene Organismen sind ständig abiotischen und biotischen Stressfaktoren ausgesetzt. Die Anpassung an diese Faktoren wird i.d.R. durch die Funktion von Phytohormonen gewährleistet, die niedermolekulare Signalmoleküle sind. Unter diesen Hormonen sind auch die Jasmonsäure (JA) und ihre Derivate, deren Zell- und Gewebe-spezifische Detektion aufgrund ihrer geringen Gehalte in pflanzlichen Zellen sehr schwierig ist. Diese Arbeit hatte zum Ziel, neue, nicht-invasive Detektionsmethoden für Jasmonate zu entwickeln und zu nutzen. Die angestrebte Methode sollte auf der Entstehung eines Fluoreszenzsignals in lebenden Zellen beruhen und spezifisch durch JA induzierbar sein. Die damit mögliche Lebendzell-Detektion von aktiver JA soll dazu beitragen, die zell-spezifische Funktion von JA in biotischen Interaktionen, aber auch in der Entwicklung von Pflanzen aufzuklären. Zusammenfassend stellen alle drei hier entwickelten Ansätze Möglichkeiten zum Zell- und Gewebe-spezifischen Nachweis von JA dar, wobei die Systeme, die auf der CSY4-vermittelten Abbau von RNA und den synthetischen JA-responsiven Promotoren beruhen, das größte Entwicklungspotential besitzen. Für alle der Methoden sind jedoch weitere Optimierungen notwendig.
As sessile organisms, plants are constantly exposed to biotic and abiotic environmental factors. Phytohormones such as jasmonic acid (JA) and its derivatives, which are low molecular weight signal molecules, control the adaptation in plants to specific conditions. Cell- and tissue-specific detection of jasmonates is still quite difficult due to their low levels within plant cells or tissues. This study aims to develop and to use a new non-invasive detection system for active jasmonates. The envisaged system should be based on the rise of fluorescent proteins in living cells appearing after specific induction by jasmonates. Such a monitoring of JA will allow getting insights to the cell specific functions of JA in biotic interactions and developmental processes. Three approaches to develop a non-invasive system for the detection of JA were followed in this study. In conclusion, three systems were developed in this study. The Csy4-mediated degradation of mRNA of reporter gene in combination with JA-mediated degradation of JAZ1-Csy4 fusion protein and the use of JAREs combined with TALE seem to be usable methods to detect JA on cell specific level. All the systems should be optimized in further.
