Abstract: NSCL-2 (neurological stem cell leukaemia) ist ein Protein aus der Gruppe der basischen Helix-Loop-Helix Transkriptionsfaktoren. Während der Embryogenese der Maus finden sich NSCL-2 Transkripte ausschließlich in neuronalen, postmitotischen Zellen in weiten Teilen des zentralen und peripheren Nervensystems. Postnatal fällt die Expression stark ab, ist in adulten Stadien jedoch noch in verschiedenen Kerngebieten des Hypothalamus zu finden. Im Rahmen dieser Arbeit wurden Untersuchungen zur Gewichtsregulation an NSCL-2 defizienten Mäusen durchgeführt. Die Tiere zeigen eine postpubertäre Obesität mit einer einhergehenden Leptinresistenz und sind infertil. Necdin, ein in postmitotischen Zellen exprimiertes Gen, wurde in in vitro und ex vivo Studien als direktes Zielgen von NSCL-2 identifiziert. Der Necdin Genlokus ist im Prader-Willi-Syndrom deletiert. Die Patienten leiden unter anderem unter Hypogonadismus und Adipositas - Symptome, die dem Phänotyp der NSCL-2 Mutanten ähneln. Durch Transaktivierungsexperimente konnten die Aktivierung des Necdinpromotors durch einen oligomeren Proteinkomplex bestehend aus NSCL-2, dem ubiqitären bHLH Faktor E12, und den Kofaktoren LMO4 und CLIM2 in neuronalen und nicht-neuronalen Zellen nachgewiesen werden. Adulte NSCL-2 defiziente Tiere zeigten eine Insulinresistenz isolierter Adipozyten, die in ob/ob x NSCL-2 -/- Doppelmutanten gemildert war. Die Insulinresistenz von NSCL-2 Einzelmutanten scheint sich jedoch auf das Fettgewebe zu beschränken, da die Blutzuckerwerte und durchgeführten Glukosetoleranztests keine Änderungen zeigten und die Serum Adiponektinspiegel sogar leicht über WT Niveau lagen. Eine weitere Auffälligkeit im weißen Fettgewebe ist die stark erhöhte Anzahl an Präadipozyten. Das im Vergleich zum Wildtyp unverändert gute Differenzierungspotential isolierter NSCL-2 -/- Präadipozyten zeigte, dass keine zellautonomen Differenzierungsblokaden vorliegen, sondern exogene Signale die Ausbildung reifer Adipozyten beeinflussen. Die Messung der Serum Resistin Konzentration ergab signifikant erhöhte Werte im Vergleich zu ob/ob Tieren. Resistin wird vom Fettgewebe sekretiert und kann unter anderem die Differenzierung von 3T3-L1 Präadipozyten inhibieren.
NSCl-2 (neurological stem cell leukemia) belongs to the protein family of basic helix-loop-helix transcription factors. During embryogenesis it is exclusively expressed in neuronal posttranscriptional cells in broad regions of the central and peripheral nervous system. Postnatal the expression declines but is maintained in central parts of the hypothalamus. One aim of this work was to analyse the central weight regulation in NSCL-2 deficient mice. The animals show adult onset of obesity, leptin resistance, and infertility. In vivo and in vitro studies identified necdin as a direct target gene of NSCL-2. Necdin is expressed in postmitotic cells and is associated with the Prader-Willi-Syndrome. For example, PWS patients suffer from hypogonadism and obesity - symptoms, which are similar to the phenotype of NSCL-2 knockout mice. With transactivation experiments in neuronal and non-neuronal cells it was shown that NSCL-2 activates the necdin promoter within an oligomeric protein complex that consists of E12, LMO4 and CLIM2. Isolated adipocytes of adult NSCL-2 deficient mice are insulin resistant, while NSCL-2 -/- x ob/ob adipocytes show decreased insulin resistance. It appears that the insulin resistance of NSCL-2 knockout mice is limited to adipose tissue, because not only serum insulin and adiponectin levels but also glucose tolerance where not changed compared to wildtype mice. The knockout animals show an increased number of preadipocytes. This cell accumulation is presumably not caused by autonomic cell malfunctions, because they show a normal in vitro differentiation potential. Therefore secondary mechanisms are hypothesised to influence maturation of adipocyten in white adipose tissue of NSCL-2 deficient mice. Serum resistin levels are increased compared to ob/ob mice. Resistin is secreted from adipose tissue and is known to inhibit the adipogenesis of the 3T3-L1 preadipocyte cell lineage.
